Validation Report Specifications
(Validation Plan): Bias and Precision
Analysis of Validation Controls
Specification: At least three validation control levels will be monitored at a low, medium, and high concentration relative to the lower and upper limits of quantitation.
• Data will represent at least three runs.
• There will be at least three replicates of each validation control at each validation control level in each run.
• Average Bias should be less than 20%.
• Average Between-Run Coefficient of Variation (BRCV) should be less than 20%.
• Average Within-Run Coefficient of Variation (WRCV) should be less than 20%.
• Validation controls that are not within quantitation limits are not evaluated for acceptability.
(Validation Plan): Calibration Model
(Quadratic, 1/X Weighted)
• Specification: A calibration model is selected during method development. Residuals for the standards will be calculated and should form a random distribution about the calibration curve.
• The coefficient of determination (R2) should be greater than 0.99.
• Standards should have an average bias measurement less than 20%.
• Standards should have an average coefficient of variation measurement less than 20%.
(Validation Plan): Sensitivity
Run to Run Robustness and Matrix Effects at [LOD Conc.] ng/mL
• Specification: Sensitivity is analyzed at the lowest calibrator level established in the calibration model.
• Samples included in this analysis represents at least three different matrices and three different runs.
• Each matrix has at least three replicates. Matrices may be pooled. The average bias measurement should be less than 20%.
Attachment D [Page 13 of 17]
• The coefficient of variation should be less than 20%. The average ion ratio deviation should be less than 20%.
• The pass percentage for each signal to noise transition should be greater than 80%.
(Validation Plan): Specificity and Selectivity
• Specification: Specificity and selectivity are measured by analyzing the chromatographic performance of the…
