About this file
This document contains a combined synopsis and solicitation for a Beckman Coulter Access System for acoustic dispensing with accessories to support SARS-CoV-2 screening efforts. The National Center for Advancing Translational Sciences (NCATS) requires a system capable of acoustic reagent dispensing without calibration, random access incubation, and multimode plate reading to enable high-throughput single agent and combination screening in a 1536-well format within their new Biosafety Level 3 laboratory. Offerors must provide the system within 60 days of award along with salient technical specifications and pricing in response to solicitation number 75N95022Q00162 by April 11, 2022. The Department of Health and Human Services National Institutes of Health National Institute on Drug Abuse will evaluate responses based on technical compliance and price to issue a firm-fixed price purchase order.

| Include a general narrative of the need of the requirement and the desired results.: ACTIV requires the acquisition of Beckman Coulter Access System OR EQUAL for Acoustic Dispensing Systems with Accessories to support the program. |
| Include a brief history of the requirement, the importance of the requirement, and the overall program objectives.: One inherent limitation of the response to the SARS-CoV2 pandemic was a readily apparent limitation of high-throughput screening capacity for biologic organisms requiring BSL3 level containment. This required the preparation of pre-spotted assay ready plates to be sent to contract research organization for screening, following their desired plate type, format, and screening in only their established assay. As such, NCATS was limited to screening SARS-CoV2 assay in 384-well format, instead of our normal 1536-well plate format, not only limiting the assay throughput but also necessitating re-tooling of compound libraries. More importantly, the assay format was restricted to only a cytopathic effect (CPE) endpoint and using only the established non-human cell type. A CPE endpoint, although a useful endpoint requires a virus inoculum and incubation period that results in direct host cell killing by the virus, limiting the utility of the assay. The cell line used in the CPE assay, a non-human monkey derived line, prevented the direct assessment of numerous drugs against SARS-CoV2 due to the inherent biological differences between this line and human cells, and in particular human lung derived cells. |
This growing demand has led to the need for additional acoustic dispensers not just for assay ready plates but to be used as a general laboratory instrument for assay development projects beyond just being used to dispense small molecules solubilized in DMSO but nearly any aqueous media. Market research has led us to learn that the only instrument available that can dispense nearly any aqueous media or non-DMSO solvent type without a specific calibration is the LabCyte Echo, meaning a user can walk up to the instrument and start dispensing WITHOUT having to have a custom and SPECIFIC calibration for every liquid type. When we were just using DMSO as our source plate solvent this wasn’t a large issue since we could calibrate for DMSO, now however we have tremendous demand to use a variety of reagent types, be it aqueous solutions, antibodies, cell media, etc. and it takes tremendous effort to create a calibration curve on the EDC and it is not always accurate and once again, is specific to that particular formulation of the reagent. For example, we have an assay that has taken literally a YEAR to get a correct calibration on the EDC to acoustically dispense cell media as the source fluid and even then there are differences from batch to batch.
To address this limitation, the National Center for Advancing Translational Sciences (NCATS) has newly established a Biosafety Level 3 (BSL3) Laboratory to support its ongoing SARS-CoV2 work. To facilitate screening, we propose to purchase (1) Access system that will permit acoustic dispense of compounds, incubation of the assay plates, dispensing of the detection reagents and reading of the plates on a multimode reader. This facilitates high-throughput single agent and combination assays to be performed in 1536-well plate format. This high-containment biological laboratory is required due to the added safety infrastructure required for propagation and handling of SARS-CoV2, including increased air handling requirements and greater security, not currently available within the 9800-laboratory complex.
Overall this project will support ongoing work for small molecule and combination screening, in silico virtual screening, nanobody/antibody design and optimization that are part of NCATS’s ongoing work to support the pandemic response. In addition, we are performing in vitro assays to support ACTIV Trace’s efforts to characterize the response of variant SARS-CoV2 viruses to current vaccine and therapeutic interventions.
