About this file
This is a Statement of Work (SOW) for the procurement of a cryostat microtome system to support prion disease research at the National Animal Disease Center (NADC) in Ames, Iowa.
The government seeks to acquire a research-grade cryostat microtome for sectioning snap-frozen animal tissues from mice, cervids, and livestock. The system must meet specific technical specifications including a temperature-controlled cryochamber with adjustable range from −10°C to −35°C, precision integrated microtome capable of producing sections 1–100 microns thick, compatibility with multiple tissue block sizes, automatic or semi-automatic cutting functionality, anti-roll plate and specimen orientation system, stainless steel or antimicrobial chamber design, and comprehensive safety features including finger guard and specimen retraction mechanisms. The equipment must include LED illumination, ergonomic design, and preferably UV decontamination or formaldehyde fumigation-compatible capabilities. Optional features include a cryobar for quick freezing samples and disposable blade holder system. The contractor must deliver and install the equipment within 45–60 days of contract award and provide on-site or remote training along with user manuals, validation documents, and warranty documentation. Evaluation factors include technical compliance with specifications, equipment reliability and ease of use, warranty length and service/support availability, delivery schedule, and biosafety and decontamination options. Delivery location is National Centers for Animal Health, National Animal Disease Center, 1920 Dayton Ave, Ames, IA 50010. The government will provide facility access for delivery, installation, and training but no additional equipment or infrastructure.

STATEMENT OF WORK (SOW)
Project Title: Procurement of Cryostat Microtome for Sectioning Snap-Frozen Animal Tissues Purpose: To support histological and molecular pathology workflows in prion disease research using precision cryosectioning of frozen tissues
1.0 Scope of Work:
This project seeks to acquire a cryostat microtome for sectioning snap-frozen mouse and large animal tissues. The system will support downstream histopathological, immunohistochemical (IHC) which is the gold standard for diagnostic evaluation of prion disease. Frozen sections allow for staining that is not always available in traditional paraffin embedded tissues. The goal is to increase efficiency, precision, and biosafety in frozen tissue preparation workflows.
2.0 Background:
Our research program routinely analyzes brain, lymphoid, and other tissues from prion-infected mice, cervids, and livestock. These tissues are snap-frozen in liquid nitrogen or dry ice/isopentane to preserve protein conformation and nucleic acids for subsequent histology, immunodetection (e.g., PrPSc deposition), and molecular assays. Cryostat-based tissue sectioning is essential for producing thin, reproducible sections from these frozen samples.
Currently, our lab does not own a dedicated cryostat system. Although the campus has a cryostat, due to the shared use of the Microscopy Service’s cryostat, it is unable to be properly deconned after TSE exposure. We are unable to use that cryostat and must purchase our own.
To support timely and consistent sample processing, we propose the purchase of a modern, research-grade cryostat with built-in safety features, temperature control, and user ergonomics.
3.0 Technical Requirements / Tasks:
The cryostat system must meet the following minimum specifications:
• Temperature-controlled cryochamber with adjustable range (e.g., −10°C to −35°C)
• Precision microtome integrated within cryochamber
• Compatible with mouse, deer, and livestock tissue block sizes
• Section thickness range: 1–100 microns
• Automatic or semi-automatic cutting functionality
• Anti-roll plate and specimen orientation system
• Stainless steel or antimicrobial chamber design for biosafety
• UV decontamination or formaldehyde fumigation-compatible (preferred)
• Safety features including finger guard, specimen retraction, and locking mechanisms
• LED illumination and ergonomic design for prolonged use
• Optional: Cryobar for quick freezing samples
• Optional: Disposable blade holder system
4.0 Government Furnished:
Access to the facility will be coordinated for equipment delivery, installation, and training. No other equipment or infrastructure will be provided by the government.
5.0 Deliverables / Schedule:
• Equipment must be delivered and installed within 45–60 days of contract award
