About this file

This is a Task Order Request for Proposals (TORP) from the Department of Health and Human Services for the manufacture and stability studies of NCATS-AAVx-001, an adeno-associated virus (AAV)-based gene therapy candidate intended for intravenous administration. The 48-month task order falls under Technical Area 3 (Biopharmaceuticals) and will be awarded as either Cost Plus Fixed Fee or Firm Fixed Price.

The contractor must develop and manufacture the gene therapy vector through multiple stages: lab scale production (1E14 vg at 10L), engineering run, manufacturing scale non-GMP material (2E15 vg at 200L+), and cGMP clinical material (1.5E16 vg in 100 vials at 1.5E14 vg/ml). Key deliverables include developing formulation buffers, analytical methods, plasmid master cell banks, and performing stability studies under ICH procedures. The work requires extensive testing at each stage including bioburden, mycoplasma, viral safety, genomic/capsid titers, impurities, and other quality attributes. The contractor must provide documentation in eCTD format and comply with cGMP regulations. Major deliverables are due within specified timeframes: assay method development report (90 days), clinical batch manufacture (210 days), batch records (30 days post-manufacture), and stability reports (30 days after each pull).
