About this file
This is a sampling protocol document developed by the University of Florida's Aquatic and Wetland Plant Science Lab, in conjunction with the Florida Fish and Wildlife Conservation Commission for the Lake Kissimmee Revegetation - Submerged Vegetation project. The protocol provides detailed instructions for collecting and preparing eelgrass samples for DNA identification of non-native aquatic plants in Florida. The sampling process requires collectors to cut leaf blades from eelgrass near the base, dry samples on paper towels to remove debris and algae, trim samples to 3-4 inches in length, and label paper envelopes with collection details including locality, coordinates, date, collector name, and contact information. Prepared samples must be placed individually in labeled envelopes and then sealed within ziplock bags filled with silica beads to maintain dryness during storage and transport. Multiple samples may be combined in a single bag provided each leaf remains in a separate envelope with unique identifying information.
Samples collected according to this protocol should be submitted to the Aquatic and Wetland Plant Science Lab at the Fort Lauderdale Research and Education Center, University of Florida, located at 3205 College Avenue, Davie, Florida 33314. The document provides contact information for Dr. Chad Keates and Dr. Lyn Gettys for any questions regarding sample collection or submission procedures. The protocol emphasizes quality control measures such as selecting fresh plant material, removing contaminants, and using adequate silica beads to ensure successful DNA extraction in subsequent laboratory analysis. No specific pricing, funding amounts, or contract award information is contained within this document.

SEARCHING FOR EELGRASS
Sampling Protocol for the Identification of Non-native Aquatic Plants in Florida
SAMPLE PREPARATION
WHAT YOU WILL NEED
- Plastic Bag
- Silica Beads
- Paper Envelopes
- Scissors
- Marker
- Towel
- Phone/GPS
STEP 1
Cut a leaf blade off of the eelgrass, starting close to the base. Try your best to avoid older plant material. The better the sample, the easier it will be to extract DNA of high quality.
STEP 2
Dry the leaf on a paper towel, ensuring to remove any debris, algae or foreign plant material from the leaf cutting.
This will reduce the chance of contami-nation in downstream processing.
STEP 3
Write the sample details onto the paper envelope with the marker. In-clude the following: general locality, coordinates, date of collection, name of collector, and contact information of collector (in case we need more info).

SAMPLE PREPARATION
STEP 5
Place the leaf cutting into the labelled envelope ensuring that you only place one leaf in the envelope. Close the envelope. You do not need to place silica beads in the envelope. Use envelopes that don’t retain water.
STEP 4
Ensure that the sample is 3 — 4 inches long using a ruler. Trim samples that are longer. Samples must not be longer than the envelope they are placed in otherwise they won’t dry properly.
STEP 6
Place the sealed envelope in a zip lock bag and fill the bag with silica beads.
Ensure that the bag is thoroughly filled so the envelope is covered. The silica beads remove moisture from bag so the more beads the better.
STEP 7
You can add multiple samples to the same bag containing silica beads. Just ensure that each leaf is in a separate envelope with unique sample infor-mation. The more samples you have the more silica beads you will need.
