About this file
This is a Single Source Justification document from the Department of State's Office of Acquisitions Management for procuring a Luminex INTELLIFLEX DR-SE Dual Reporter system with Belysa® software. The procurement is for the WRAIR-AFRIMS Virology unit's field-based surveillance projects in Thailand, specifically for projects studying dengue, febrile illnesses, and respiratory pathogens. The system requires unique capabilities including dual reporter technology, high multiplexing capacity (up to 500 analytes per sample), advanced automation features, and specific technical requirements such as simultaneous detection using two laser wavelengths and compatibility with 96- and 384-well plates.
The recommended system configuration includes the xMAP® INTELLIFLEX DR-SE RUO Instrument, Belysa® analysis program, calibration kit, sheath fluid, sample probe accessories, heater block, magnetic separator, and other supporting equipment. Market research justified the single-source procurement, emphasizing that only this specific system with Belysa® software meets the agency's critical needs for advanced immunoassay workflows, including features like advanced curve-fitting, batch quality control, parallelism testing, and automated outlier detection. The procurement includes installation, user training, and a one-year warranty, with the total estimated value redacted in the document.

Text version
• Dynamic Range and Sensitivity: Typical dynamic range of 5.5 logs for the 532 nm reporter and 4.5 logs for the 405 nm reporter, with sensitivity reaching sub-picogram levels depending on assay kits.
• Analytical Software
Analytical Software Requirement: Belysa® Immunoassay Software To fully realize the value of the Luminex INTELLIFLEX DR-SE system, the required procurement must include Belysa® Immunoassay Curve Fitting Software. Belysa® is optimized for high-plex immunoassay data, offering:
• Advanced curve-fitting models (5PL, 4PL, linear, spline)
• Built-in plate-to-plate and multi-analyte QC monitoring
• Outlier flagging, standard curve comparability, and parallelism testing
• Customizable acceptance rules for batch-level data integrity
• Exportable and traceable data audit trails for reproducibility
These capabilities are critical for surveillance programs relying on longitudinal consistency, multi-site data comparability, and low-volume precision. Other analysis options provided do not provide essential features such as:
• Parallelism and spike recovery analysis
• Batch QC rule-setting
• Automated outlier detection and customizable result filtering
• Integration with lab-developed tests or non-ProcartaPlex assay kits
Recommended System Configuration and Items To meet these technical and operational requirements, the laboratory seeks to procure the Luminex INTELLIFLEX DR-SE Dual Reporter system bundled with Belysa® software, including the following essential components:
• xMAP® INTELLIFLEX DR-SE RUO Instrument 1 EA
• Calibration and Performance Verification Kit 1 Set
• Sheath Fluid PLUS, 20 L 1 EA
• Belysa analysis program 1 EA
• Sample probe needles 3 EA
• Sample Probe Height Adjustment Kit 1 EA
• Off-plate reagent block 1 EA
• Keyboard and Mouse 1 Set
• Heater Block for 96 and 384-Well Microtiter Plates 1 Set
• Empty Cubitainer for Waste 1 EA
• Handheld Magnetic Separator Block 1 EA
• Uninterruptible Power Supply (UPS) 1 EA
• User guide manual 1 EA
• Installation and user training provided by manufacturer-trained personnel until users are proficient
• One-year warranty under normal use conditions
Total Estimated Value The estimated total value for this procurement is inclusive of all equipment, software, accessories, warranty, and training.
