About this file

This statement of work and related solicitation document outline requirements for six dCas9-KRAB cell lines to support genome-wide CRISPR interference screening conducted by the National Center for Advanced Translational Sciences Functional Genomics Lab. Cellecta, Inc. will be directly awarded a contract to generate the six cell lines with inducible targeted gene inhibitions within twelve weeks of order receipt. The cell lines will be delivered to the National Institutes of Health National Center for Advanced Translational Sciences for use in further screening and downstream biological analysis to identify promising gene targets and gain groundbreaking insights. This non-competitive procurement is being conducted by the National Institute on Drug Abuse on behalf of NCATS pursuant to Federal Acquisition Regulation Part 13 authorities for simplified acquisition of commercial items below the threshold.

STATEMENT OF WORK (SERVICES)

(SOW)

GENERAL INFORMATION

Title of Project: High-throughput CRISPR Interference (CRISPRi) screening to understand the pathobiology of Acral lentiginous melanoma (ALM) and to determine genes required for ALM survival.

Statement of Need and Purpose: The purpose of this procurement is the acquisition of six dCas9-KRAB cell lines.

Background Information and Objective: The National Institutes of Health has established an RNAi/CRIPSR Cas9 screening facility to provide intramural researchers with access to the equipment and expertise necessary to conduct genome-wide RNAi/ CRIPSR Cas9 screens in mammalian cells. This facility works as a collaborative resource for doing RNAi/ CRIPSR Cas9 assay development, screening, and initial follow-up. The facility works with investigators to develop and optimize their assays for screening, conducts the screens and related informatics analyses, and then performs follow-up screens on promising leads. Further work is done with the PI to do the downstream biology required to get the quality publications and the kind of groundbreaking results needed. This request is required for this facility to develop a flexible functional genomics platform that offers complementary and orthogonal RNAi and CRISPR Cas9 screens to NIH intramural investigators.

The required dCas9 cell lines will be used to repress the target gene expression when applied in a CRISPRi screen. This item is needed to facilitate the RNAi/ CRIPSR Cas9 screening program.

Period of Performance: 4/3/2024- 6/26/2024 or 12 weeks after receipt of the order
