Statement of Work (SOW)

Project: Production of Genetically Engineered Cells and Livestock Species.

GENERAL INFORMATION

1.0 Scope of Work: The overall goal is to produce genetically engineered cells and livestock species that have improved agricultural traits. Genetic modifications will be produced by electroporation and lipofection and then single cells need to be grown from single cells into many cells. This is called single colony formation. This is hard to isolate single cells and then allow these cells to grow without other cells, but it is a critical part of generating many cells all containing the same edit for phenotyping analysis.

2.0 Background: Research is being conducted to develop pigs, poultry and cows with improved traits of interest for the livestock industry by using genome editing techniques.

The scientists will use genomic information to predict DNA sequence changes that will cause beneficial changes in livestock animal production (improved nutrition, improved digestibility, overall animal health and welfare: disease resistance, etc.) and to test those changes, in vitro phenotyping of cells and organoids will be completed. After genome modifications are created in the cells the change in phenotype will be validated in vitro before moving forward to produce a genome edited animal. Once we have determined the DNA sequence is beneficial to the trait of interest, we will generate an animal(s) with the same DNA sequence. The Cell Raft AIR system request will be used to sort individual cells into microwells that share the same medium and proximity to other cells creating a more suitable environment for growing single colonies. This microwell technology will allow hard to grow single cells to develop into single colonies to ensure we are able to recover monoclonal cell cultures that have the same DNA modifications. This will allow us to determine if the DNA modification is associated with a positive trait or not.

CONTRACTOR REQUIREMENTS

3.0 Technical Requirements/Tasks:

• Must be able to automatically sort individual cells into microwells that maintain close proximity and share the same medium to other cells in the plate.

• Must be able to provide time-course imaging of all wells for clonal verification and cell health monitoring.

• Must be able to sort, culture, and image up to 40,000 microwells at a time.

• Must be able to automatically move cells from microwell system to a 96 well plate without disturbing the colony.

• Must have the ability to sort fluorescently labeled cells.

• Must include installation, training and calibration Array.

CellRaft AIR System includes:

• AIR Systems Accessory Kit - CytoSort® Array Adapter Plate, PCR Tube Adapter Plate, Standard

• Collection Wand (2), and PCR Tube Collection Wand (2)

• AIR System AIO Computer - Power cord (1.5m), wireless keyboard and mouse, two (2) USB 3.0 cables

• Pre-installed with AIR Software. System computer has a 10th Generation Intel Core processor

• Window 10 Pro installed, a 16GB Ram, 256GB solid state drive, Wi-Fi and Ethernet connectivity.

• 0.5 TB portable solid-state drive with "Off-the-AIR" Software

4.0 Government Furnished: Not applicable

5.0 Deliverables / Schedule: Must provide 1-year extended warranty. Must be delivered 120 days after award. The equipment must be delivered to:
