Minimum Specifications Project: Purchase One Atomic Force Microscope
GENERAL INFORMATION
1.0 Scope of Work:
An atomic force microscope is required to visualize misfolded prion fibrils, the causative agents of Scrapie and Chronic Wasting Disease, to support the objective of understanding the pathogenesis of transmissible spongiform encephalopathies (TSEs) through studying prion misfolding and fibrilization events.
The research goal is to compare the morphology of prion fibrils from chronic wasting disease, classic scrapie and atypical scrapie, compare different fibril populations within each disease type, and measure fibril length and diameter using atomic force microscopy. The atomic force microscope can then be used to visualize the effects of various remediation agents such as biochar on prion fibril stability and fibril complexes and visualize the binding of prion fibrils to these agents.
2.0 Background:
The prion research unit does not currently have access to an atomic force microscope and standard microscopes lack the necessary resolution to visualize individual prion fibrils. The atomic force microscope must be a complete system that includes a high-resolution atomic force microscope with a small footprint that can visualize atomic-scale surface features, control station with software, and image analysis software. The system must be user-friendly with the potential to upgrade its capabilities in the future. Installation, training and application support are required.
CONTRACTOR REQUIREMENTS
3.0 Technical Requirements/Tasks
1. Atomic force microscope must be high resolution research grade instrument
2. Atomic force microscope must have a minimum 50 micron XY 17 micron Z scanner
3. Atomic force microscope must have precision Z motorized approach
4. Atomic force microscope must have precision XY stage with micrometers
5. Atomic force microscope must have universal probe holder
6. Atomic force microscope must have minimum 28-bit scanning
7. Atomic force microscope must have a video optical microscope with a minimum
45X magnification (20 micron resolution) and be capable of 400X magnification (2 micron resolution)
8. Atomic force microscope must be supplied with control software, computer and all other computer accessories needed to run instrument
9. Atomic force microscope must have no more than a 36 x 36 inch footprint
10. Atomic force microscope must come supplied with acoustic and structural isolation cabinet and cabinet base
11. Atomic force microscope must include installation, on-site training and application support
12. Atomic force microscope must support future technical upgrades
4.0 Government Furnished:
Instrument will be located in USDA, NCAH, Bldg 20, Room 2630, 1920 Dayton
Ave., Ames, IA 50010. Installation and training will be scheduled to take place during regular working hours, except all federal holidays.
5.0 Deliverables/Schedule
Delivery and installation of the equipment should occur as soon as the instrument becomes available
6.0 Evaluation Factors
Evaluate based on lowest price technically acceptable
