Statement of Work

MALDI-TOF Mass Spectrometry System

GENERAL INFORMATION

1.0 Scope of Work: The United States Department of Agriculture (USDA) National

Animal Disease Center in Ames, IA has a need for a MALDI-TOF System to identify microorganisms using MALDI-TOF (Matrix-Assisted Laser Desorption /

Ionization Time-of-Flight) mass spectrometry to determine the unique proteomic fingerprint of an organism by measuring the highly abundant proteins that are found in all microorganisms and uses their specific differences for identification.

2.0 Background: MALDI-TOF technology has been adopted in many microbiology laboratories worldwide, for reliable, fast and efficient identification of a wide range of gram-negative and gram-positive bacteria, yeasts and molds, by an easy to operate, yet powerful benchtop system. USDA researchers have identified this technology as a tool to support current research. Raw data and reference databases are needed to interpret results, so a complete system is required for this need.

CONTRACTOR REQUIREMENTS

3.0 Technical Requirements/Tasks:

1. MALDI-TOF system must include that reference library entries in the MALDI system are stored and capture the biological variability of an organism. The system must compare unknowns to the library using a pattern-matching approach.

This includes peak positions and intensities, ensuring the highest possible levels of accuracy and reproducibility across the complete range of microorganisms.

2. The database must be continuously updated, include a minimum of 4000 species, and can be edited to include novel strains identified by the user.

3. Raw MALDI-TOF data must be exportable.

4. MALDI-TOF must use a minimum 200 Hz laser that can be run in both positive and negative ion detection modes.

5. Sample plate must be capable of holding a minimum 95 samples and must be capable of analysis time of less than six (6) minutes.

6. System must be capable of a 0-500,000 Dalton mass range

7. Instrument dimensions shall not exceed 20’’ x 30’’ x 45’’ (L x W x H) and 175 pounds.

8. Operational conditions shall not exceed 60 decibels.

9. System must include an oil-free membrane pre-vacuum pump

10. Quote shall include operating software, delivery, onsite installation and onsite training.

11. Qty: 1 complete system.

4.0 Deliverables/Schedule: Shall be delivered onsite within 30 days of date of award.

5.0 Evaluation Factors: Technical acceptability and price with technical specification more important than price

6.0 Place of Delivery: USDA, 1920 Dayton Avenue, Bldg 24, Ames, IA 50010
