About this file

This Performance Work Statement (PWS) outlines requirements for RNA sequencing services of wheat RNA samples for the USDA Agricultural Research Service. The vendor must analyze two sets of wheat RNA samples: Set I containing 72 samples and Set II containing 60 samples.

The technical requirements specify that RNA samples will be provided at ≥100 ng quantity with a concentration of ≥10 ng/uL and volume ≥10 uL, plus 5 uL per sample for quality control. The service includes RNA sample quality control evaluation, mRNA library preparation with Poly-A enrichment, and 150 bp paired-end reads generating 6 Gb raw data per sample with Q30 ≥85%. The vendor must perform data quality control to remove adapter sequences and low-quality reads, and return the data within 21 days of sample submission via FTP file transfer.

RNA Sequencing of Wheat RNA Samples: 72 samples and 60 samples

Performance Work Statement (PWS)

The vendor will provide RNA sequence data from two sets of wheat RNA samples provided by USDA-ARS. RNA samples to be provided will be:

• ≥ 100 ng (quantity) at a concentration of ≥ 10 ng/uL and volume ≥ 10 uL

• An additional 5 uL per sample will be provided for RNA sample quality control evaluation

Set I: 72 Samples

Set II: 60 Samples

This service will include:

• RNA sample quality control evaluation to verify RNA integrity

• mRNA library preparation (Poly-A enrichment)

• 150 bp paired-end reads o 6 Gb raw data per sample o Q30 ≥ 85%

• Data quality control to remove reads containing adapter sequence or with low quality.

• Data returned within 21 days of sample submission via FTP file transfer
