About this file
This is a Single Source Justification Form (FWS Form 3-0034) submitted by the U.S. Fish and Wildlife Service for a sole source acquisition under the Simplified Acquisition Threshold (SAT).
The U.S. Fish and Wildlife Service requires genetic data collection services from Chinook salmon at Coleman National Fish Hatchery using GTSeq methodology. The data will be used by state agencies (California Department of Fish and Wildlife, California Department of Water Resources), tribal entities (Columbia River Inter-Tribal Fish Commission), private sector partners (Cramer Fish Sciences), and federal agencies (NOAA Fisheries). Market research confirmed that all GTSeq data collection protocols rely exclusively on instruments manufactured by Illumina, establishing a single source basis for acquisition. The contractor's qualifications were validated through peer-reviewed scientific literature demonstrating that GTSeq protocols employing Illumina sequencers meet the project requirements. Fair and reasonable pricing was determined based on market research of similar past awards. The MiSeq i100 Plus System is identified as the Illumina instrument best suited to meet the stated needs. Program Officer Steven Dyer recommended sole source acquisition procedures and certified the justification as accurate on April 24, 2026. Contracting Officer Melissa Niemi approved the use of sole source acquisition procedures under FAR Part 13 on June 2, 2026.

1. Description of Need
3. Other Capable Sources
2. Market Research Conducted
Pursuant to FAR subpart 13.106-1(b)(1)(i) and (ii), for purchases not exceeding the Simplified Acquisition Threshold (SAT), Contracting Officers may solicit from only one source if the circumstances deem only one source is reasonably available (e.g., urgency, exclusive licensing agreements, brand name, or industrial mobilization).
Instructions: The information in the text boxes are provided as sample language or to indicate a needed fill-in. Sample language should be removed from the final document. All text boxes require information and shall not be left blank.

| Text2: After speaking with colleagues and the entities listed above, an extensive online search of the scientific literature was performed.. Every one of the protocols described for collection of GTSeq data relies on instruments built and sold by Illumina. | |
| Text1: We need to collect genetic data from Chinook salmon from Coleman National Fish Hatchery, which can then be used by State (e.g., California Department of Fish and Wildlife, California Department of Water Resources), Tribal (e.g., Columbia River Inter-Tribal Fish Commision) private sector (e.g., Cramer Fish Sciences), and Federal (e.g., NOAA Fisheries) partners. In order to meet these criteria, the genetic data needs to be collected using the GTSeq methodology (published here: https://onlinelibrary.wiley.com/doi/full/10.1111/1755-0998.12357). | |
| Text4: The GTSeq chemistry was designed around proprietary Illumina technology. In order for us to generate data that can be integrated with our partner's data, we thus need an Illumina instrument, and the MiSeq i100 Plus System is the Illumina instrument which will best meet our needs. | |
| Text6: Pricing is determined to be fair and reasonable based on market research of past similar awards. | |
