Version 11-30-2016-1 FAR Part 11—Describing Agency Needs
STATEMENT OF WORK (SERVICES)
(SOW)
GENERAL INFORMATION
Title of Project:
Developing selective small-molecule modulators of PIP4Kgamma to better understand its biology and evaluate its therapeutic potential.
Statement of Need and Purpose:
This purchase order supports pharmacokinetic (PK) studies of the PIP4Kgamma lead compound NCGC00592266 in mice following intraperitoneal (IP) administration at 10 mg/kg once daily for 7 consecutive days (QD×7). Samples will be collected from plasma, brain, liver, and pancreas at the following time points: 5 min, 15 min, 30 min, 1 h, 2 h, 4 h, 8 h, 12 h, and 24 h (n=3 mice per time point).
Drug concentrations will be measured in plasma and tissues to evaluate systemic exposure, tissue distribution, and potential accumulation following repeated dosing. These data will support dose selection, assess tissue exposure and safety considerations, and facilitate advancement of the compound into preclinical development.
Background Information and Objective:
The National Institutes of Health (NIH) is the nation’s leading medical research agency and the primary Federal agency whose mission is to seek fundamental knowledge about the nature and behavior of living systems and the application of that knowledge to enhance health, lengthen life, and reduce illness and disability, conducting, supporting and making medical discoveries that improve people’s health and save lives.
The National Center for Advancing Translational Sciences (NCATS, Early Translation Branch) is a part of the National Institutes of Health (NIH), whose mission it is to catalyze the generation of innovative methods and technologies that will enhance the development, testing and implementation of diagnostics and therapeutics across a wide range of human diseases and conditions.
PIP4Kgamma (phosphatidylinositol-5-phosphate 4-kinase gamma) is a lipid kinase involved in phosphoinositide signaling, regulating cellular processes such as membrane trafficking, autophagy, and metabolic homeostasis. Unlike other PIP4K isoforms, PIP4Kgamma is enriched in the central nervous system, making it of interest for neurological and metabolic disorders. The project focuses on developing selective small-molecule modulators of PIP4Kgamma to better understand its biology and evaluate its therapeutic potential. Early studies suggest that modulation of PIP4Kgamma can impact pathways linked to neurodegeneration, lysosomal function, and energy balance. The lead compound (e.g., NCGC00592266) has been identified through medicinal chemistry optimization and is being advanced to characterize its pharmacokinetics, tissue distribution (including brain penetration), and in vivo activity to support further preclinical development.
Period of Performance:
Documentation of results and presentation of these results to NCATS via email and biweekly conference calls by vendor over a 1 month period after receipt of PO.
