2. Technical Background and Contractual History 7.105 (a) (1)

This is a new requirement

The 32-plex 10x Genomics 5' GEX/TCR/BCR kit is essential for the proposed studies of B- and T-cell clonality in Sjögren’s disease salivary glands. The project requires simultaneous single-cell characterization of transcriptomes and paired antigen receptor repertoires to identify clonally expanded lymphocyte populations and define their functional states.

No alternative method available in the laboratory can provide this integrated analysis with equivalent single-cell resolution, throughput, and sample multiplexing capacity.

The 32-plex configuration is necessary to reduce technical variation across samples, maximize use of limited patient specimens, and enable statistically meaningful comparison of disease and control groups.

These studies will provide critical mechanistic insight into the role of adaptive immune clonality in SjD pathogenesis and are directly aligned with the project’s goals of identifying disease-driving cell populations and informing future biomarker and therapeutic development.

Not Applicable

Brand Name Requirement (A brand name justification is attached)

Sole Source Requirement (A sole source justification is attached)

4. Compatibility or Performance Characteristics for Supply of Service 7.105 (a) (4)

Required Features & Benefits of the acquisition:

We require these reagents for high-resolution, single-cell analysis of a novel human minor salivary gland epithelial 3D model system. We require the 5’ GEX/TCR/BCR Immune Profiling Kit from 10x Genomics.

This platform provides the essential capability to perform single-cell RNA sequencing (scRNA-seq) cryopreserved samples, while maintaining high transcript fidelity and cell recovery rates. The kit's high-throughput, scalable, and standardized format allows reproducibility and data harmonization across multiple samples and cohorts.

Required capabilities/performance characteristics/standards

The required kit must support high-throughput single-cell 5' gene expression profiling together with paired V(D)J immune repertoire sequencing for both T-cell receptors (TCR) and B-cell receptors (BCR) from the same cells, with sample multiplexing up to 32 samples in a single workflow. It must provide sufficient sensitivity, cell recovery, and library compatibility to identify clonally expanded B- and T-cell populations and directly link clonotype to transcriptional state.

Why these are necessary and how they relate to the need

These capabilities are necessary because the study objective is to define B- and T-cell clonality in Sjögren’s disease while simultaneously determining the functional state of those same lymphocytes. Bulk repertoire methods or standard expression assays cannot connect clonal expansion with single-cell phenotype. The 32-plex multiplexing capacity is directly related to the need to analyze multiple patient and control samples efficiently, reduce batch effects, conserve limited clinical specimens, and enable rigorous cross-sample comparison. Together, these performance characteristics are essential to generate the integrated single-cell immune profiling data required to identify disease-associated clonal populations in SjD.

Compatibility/Performance Requirements (describe compatibility or interfaces with current and/or future systems/programs, describe any compatibility or performance risks/issues related to the requirement, describe any clearances such as IT security affiliated with this requirement, describe any government furnished equipment affiliated with this requirement)

This system is compatible with Chromium X instrument’s proprietary microfluidics, barcoding chemistry, and software that is widely used throughout the NIH and NIDCR.
