STATEMENT OF NEED AND REQUIREMENTS

Direct Electron Detector suitable for NIEHS Talos Arctica cryogenic electron microscope

NIEHS has deployed a productive program in cryo-electron microscopy (cryo-EM), a powerful set of emerging technologies capable of solving the structure of complex macromolecular assemblies from small quantities of purified material and/or in their biological environment. The Cryo-EM Core, working in the context of the Molecular Microscopy Consortium, a strategic alliance with Duke and UNC, has contributed to the adoption of the methodology by over two dozen research groups. These efforts resulted in close to a hundred publications in high-profile journals. The Cryo-EM core has also established intramural collaborations, most notably with several groups in the intramural program (NCATS, NCI, NIA and NIDA) in the context of structural biology research related to, among other important topics, the COVID-19 epidemic.

The first high resolution cryo-EM capable instrument in the region, a Thermo-Fisher (TFS) Talos Arctica cryogenic transmission electron microscope (cryoTEM) equipped with a Gatan K2 Summit Direct Electron Detector (DED) was installed at NIEHS seven years ago. The DED detector has now reached the end of its service cycle and needs to be replaced to ensure the continuity of our operation. Thus, the Cryo-EM

Core requires a Direct Electron Detector to replace the Gatan K2 Summit installed on the Thermo Fisher Talos Arctica cryogenic electron microscope.

The K2 is becoming obsolete after seven years of excellent service. The new detector should meet the capabilities of the K2, remain compatible with the microscope’s hardware and software as well as with the rest of the data collection and image processing pipelines. Expansion of abilities to include micro-electron diffraction studies on protein crystals is highly desirable.

The DED must meet the following criteria:

• high-performance direct detection counting capable of collecting single-particle, tomography, and electron diffraction data.

• Radiation resistant for electron diffraction data collection without a beam stop.

• Full support of TEM operation from 80 – 200 keV.

• Field-of-view must be 4K x 4K pixels or larger in counting mode with oversampling capabilities.

• Correlated-double sampling mode to reduce noise.

• The sensor readout speed must be ≥1,000 fps and electron counting must be performed in real-time at the sensor readout speed.

• Optional inline motion correction.

• DQE must be ~0.9 at peak and ~0.4 at Nyquist.

ATTACHMENT 1

• The DED should be compatible with open-source automated data collection software packages for acquiring single-particle, tomography, and electron diffraction data.

• Multiple data formats including hdf5, mrc, and tiff should be supported.

• Camera hardware and computer providing over 10 TB RAID storage and 10 G network cards should be included.
