About this file
This document is an amendment to a request for proposals for a virology core laboratory. The amendment provides responses to questions from potential offerors to clarify requirements. The laboratory will be required to conduct approximately 4,000 assays annually to detect simian immunodeficiency virus (SIV) or simian-human immunodeficiency virus (SHIV) viral loads in addition to 200 assays for assay optimization and 200 assays for development of an assay to detect cell-associated SIV DNA. The budgets for the assays under Options 7 through 13 should include materials and supplies costs for approximately 350 SIV viral RNA assays while the effort for those options should also be budgeted. The proposal due date remains December 7, 2023 at 3:00 PM Eastern Time and the solicitation number is 75N93023R00017 issued by the National Institute of Allergy and Infectious Diseases of the National Institutes of Health within the Department of Health and Human Services.

Please note that we do NOT intend to extend the due date for submission of proposals as a result of this amendment. The date specified for receipt of proposals remains unchanged.
Offerors must acknowledge receipt of this Amendment by identifying the amendment number and the date of the amendment on each copy of the submitted proposal. Failure to receive your acknowledgement may result in the rejection of your proposal. Except as provided herein, all terms and conditions of the solicitation remain unchanged and in full force and effect.
The purpose of this Amendment is to provide responses to questions from potential offerors, regarding the solicitation.
RESPONSES TO QUESTIONS FROM POTENTIAL OFFERORS:
1. How many assays should be budgeted for SIV or SHIV vs HIV? Is it anticipated that the majority of the expected testing would be for detection of SIV or SHIV viral load?
Response: Yes, please budget for assays that will detect SIV or SHIV.

AMENDMENT #2 RFP No. 75N93023R00017 2
2. How many assays should be included for assay optimization and assay development in the budget?
Response: For cost assumption purposes, plan to annually conduct 200 assays to optimize your current QPCR assay to increase the sensitivity. This is in addition to the budget for the 4,000 assays to detect SIV viral RNA as stated above.
3. Should the number of assays budgeted for assay development and assay optimization be subtracted from the 4000 assays requested each year? Or, is costs for assay development and optimization separate from the 4000 assays?
Response: For cost assumption purposes, plan to annually conduct 200 assays for the development of an assay that can detect cell associated SIV DNAs. This is in addition to the budget for the 4,000 assays to detect SIV viral RNA as stated above.
4. How many tissue samples are anticipated and should quantitative detection of tissue samples be subtracted from the 4000 assay listed in the cost assumptions?
Response: For cost assumption purposes, do not budget for detecting nucleic acid in tissue samples.
5. Can you confirm the budget for options 7-13 should only include effort?
Response: For Options 7-13, budget materials and supplies cost for approximately 350
SIV viral RNA assays.
