Background and Requirement
The U.S. Food and Drug Administration (FDA), Center for Drug Evaluation and Research (CDER), Office of Clinical Pharmacology (OCP), Division of Applied Regulatory Science (DARS) has a need for a 3D multi-electrode array (MEA) platform to measure electrical signals generated from organoids produced by human induced pluripotent stem cell (iPSC) to address mechanisms-of-drug action. Organoids are 3D model systems to study neural circuit dynamics in vitro. Unlike the 2D monolayer cultures, organoids self-organize to form layered neuronal architectures with glial populations and emergent synaptic connectivity that more closely reflect in vivo brain tissue organization.
The neuronal populations underlying physiologically and pathologically relevant circuit functions are not confined to a single plane. Rather, they are spatially distributed across the tissue depth, forming circuit nodes that communicate across three dimensions. A single recording plane performed by 2D MEA platform therefore is inadequate as it either captures a small fraction of the relevant network activity on the surface or misses deep-layer activities not obvious on the surface. To best assess drug effects in brain organoids, the 3D MEA platform is better suited as it uses distributing electrodes throughout the tissue volume to enable simultaneous recording across depth. Electrophysiological outcomes including spontaneous firing rate, network burst frequency and duration, inter-burst interval regularity, synchrony index, local field potential power spectra, and spatiotemporal burst propagation patterns collectively reflect network functional state and pharmacological responsiveness. Tracking these measures across tissue depth could reveal how and what types of drug-induced activity changes initiate and propagate through the neural circuits.
This procurement is consistent with FDA’s Roadmap to Reducing Animal Testing in Preclinical Safety Studies (April 2025) and the draft guidance General Considerations for the Use of New Approach Methodologies in Drug Development (March 2026). The Contractor shall provide a complete, fully integrated 3D MEA instrument system, including all required accessories, software, computing hardware, installation, calibration, and training as specified herein.
The 3D MEA system will be used to:
1. Record network-level electrophysiological activity from human iPSC-derived brain organoids containing physiologically relevant neuronal populations, capturing signals from within the three-dimensional tissue volume rather than solely from the organoid surface.
2. Resolve depth-specific and spatially distributed neural firing patterns across distinct regions of the organoid, enabling identification of circuit-level changes capable of penetrating to maximum depths of 200 μm to ensure comprehensive spatial coverage of electrophysiological activity throughout the full tissue volume that would be undetectable with surface-only or 2D electrode configurations.
3. Characterize drug-induced perturbations in neural circuits, with spatial resolution sufficient to distinguish region- and layer-specific responses within the organoid tissue volume.
The Brand Name or Equal System shall meet or exceed the following minimum technical specifications:
MINIMUM TECHNICAL SPECIFICATIONS
Volumetric Recording System: System shall
1. Record electrophysiological signals from within the full three-dimensional volume of iPSC-derived brain organoids, with electrodes distributed across multiple depths to capture activity from inside the tissue rather than just the surface.
2. Utilize adjustable probes/electrodes that allow for flexible positioning and can be extended to cover the whole depth of the organoid, penetrating to depths up to 200 μm to ensure comprehensive spatial coverage of electrophysiological activity throughout the tissue volume.
3. Provide sufficient electrode density and spatial coverage to distinguish firing patterns and network dynamics at different depths and regions of the organoid tissue volume.
4. Maintain a minimum sampling rate of 10 kHz/electrode across all recording depths simultaneously, with sufficient resolution to faithfully capture both action potentials and local field potentials.
5. Feature an integrated environmental chamber that maintains stable temperature, CO₂, and humidity throughout extended recording sessions to sustain organoid viability.
6. Support compatibility with organoid-specific 3D MEA consumables designed for volumetric recording, including custom-designed 1mL single microwell recording plates and microfabricated electrophysiology probes.
Scanning and Analysis Software Capabilities: Instrument system shall:
1. Provide data acquisition and analytical modes that are fully computer-controlled and user-programmable.

Full Text Provisions The provision at FAR 52.212-1 Instructions to Offerors - Commercial Products and Commercial Services (DEVIATION)(RFO Nov 2025) applies to this solicitation. The following addenda apply:
Quoters shall provide sufficient technical information necessary for the government to conclusively determine that the offered service/equipment meets all the technical requirements herein, to include descriptive material, literature, brochures and other information corresponding to each minimum technical requirement which demonstrates the capabilities of the offered system. The Government is not responsible for locating or securing any information which is not identified in the quote; however, the Government reserves the right to obtain information for use in the evaluation from any sources including sources outside of the Government. The Government reserves the right to request additional information at any time.
Technical Acceptability: Quoter must provide sufficient technical information for the Government to conclusively determine that the quoted product meets or exceeds the minimum requirements identified above. In addition to identifying manufacturer, make, and model of quoted products, it is incumbent of quoters that they unequivocally demonstrate that quoted products meet the requirements herein through the submission of technical specifications, descriptive material, scientific literature, brochures, scientific publications where proposed solution has been used for same or similar purposes, and other information which demonstrates the capability of the quoted equipment. Quoters shall address the technical requirements identified above, as well as provide detailed information on the item(s) in the solicitation.
To be considered for award, quotes of “equal” products, including “equal” products of the sole source manufacturer shall meet the salient physical, functional, or performance characteristic specified in this solicitation; clearly identify the item by brand name, if any; and make or model number. “Equal” quotes shall include a narrative describing how the offer meets each salient characteristic as well as a demonstration of how each salient characteristic is fully met.
The Quoter shall submit a written Accessibility Conformance Report (ACR) for each item not in compliance with 508 standards identified in clause 352.239-79, describing how the item(s) will fully address the accessibility requirements outlined in the solicitation; and a description of the evaluation methods the offeror will use to validate for conformance to the Revised 508 Standards. The ACR should be based on the Voluntary Product Accessibility Template Version 2.0 (MS Word) provided by the Industry Technology Industry Council (ITIC).
http://www.itic.org/policy/accessibility
For this requirement, it is anticipated the 508 requirement will only be applicable to invoices and service records/reports.
NOTE: Submission of the ACR is required for documentation purposes. In accordance with agency purchasing procedures, solutions which do not conform fully to the applicable 508 standards may still be purchased by FDA if the FDA Requiring Office obtains the appropriate approval. In order to obtain approval, the FDA must have an ACR from the vendor documenting the level of conformance of the items being purchased.
Letters of Supply Quoters who are authorized resellers of an offered item, and not the manufacturer, shall provide a copy of their letter of supply or other documentation substantiating their relationship as an authorized reseller of the proposed manufacturer’s product. The documentation shall be on the Original Equipment Manufacturer (OEM) letterhead and include contact information for the individual who has signed the letter.
Price: The price(s) proposed must be detailed and shall represent the quoter’s response to the schedule of supplies/services above. Quoters are also encouraged to submit a vendor quote with this document.
The option year service agreement periods will not be funded at time of award. If exercised, funding will be obligated individually each year for the option period exercised via contract modification; the option periods shall be priced accordingly.
Include the firm’s Unique Entity ID (UEI) number with quote. Proprietary and/or confidential information shall be clearly marked.
The government is not responsible for locating or securing any information which is not identified in the proposal; the Government reserves the right to obtain information for use in the evaluation from any and all sources including sources outside of the Government.
The Contractor’s quote shall also clearly show that the Contractor has selected the appropriate certification under FAR 52.222-48 (see below).
Note: This solicitation is being issued under the premise that the offeror will certify that the combination of equipment maintenance/service it will propose to fulfill this requirement is exempt from the Service Contract Act per the conditions set forth in FAR 52.222-48 & FAR 22.1003-4(c). If the Contractor certifies that the company is exempt under the terms and conditions of 52.222-48, then clause 52.222-51 will be applicable to the resulting order. In the event that a contractor does not make this certification, the traditional Service Contract Act clauses 52.222-41, and 52.222-43 shall be included in the resulting order award as Wage Determination WD 2015-4269 (Rev. 33), date of last revision: 07/30/2026, which can also be accessed here: https://sam.gov/wage-determination/2015- 4269/33

When submitting proposals via email, DO NOT include .exe, .mso, or any other executable file types that could potentially trigger email security protections (i.e. email blocks, quarantine). If the offeror or applicant submits documents that contain macros, macro referenced files, and/or executable files, the Government will not be able to view or open such documents and the submission will be considered non-responsive to the solicitation. No additional time will be given to an offeror or applicant to correct the document submission and the Government will not inform the offeror or applicant that their submission is non-responsive prior to award. It is the offeror’s or applicant’s responsibility to ensure all electronic documents are submitted without the use of macros.
The provision at 52.212-2 Evaluation-Commercial Items (DEVIATION)(RFO Nov 2025) is applicable to this solicitation.
The following subparagraph (a) factors apply:
(a) The Government intends to award a contract resulting from this solicitation to the lowest priced technically acceptable responsible quoter/offeror whose quote, conforming to the solicitation, will be most advantageous to the Government, price and other factors considered. Quotes will be evaluated on their ability to meet the requirements provided herein. The lowest priced quote will be evaluated first. If the lowest priced quote is not technically acceptable, the next lowest priced quote will be evaluated and so on until a technically acceptable quote is determined.
Technical acceptability will be determined by review of information submitted by the quoter which must provide sufficient technical information necessary for the Government to conclusively determine that the quoted products meet or exceed the technical requirements identified herein.
The Government’s determination of technical acceptability in no way relinquishes the Awardee’s contractual obligation to ensure the quoted products meet the FDA’s stated need.
(b) Options. The Government will evaluate offers for award purposes by adding the total price for all options to the total price for the basic requirement. The Government may determine that an offer is unacceptable if the option prices are significantly unbalanced. Evaluation of options shall not obligate the Government to exercise the option(s).
(c) A written notice of award or acceptance of a quote, mailed or otherwise furnished to the successful quoter within the time for acceptance specified in the quote, shall result in a binding contract without further action by either party. Before the quoter’s specified expiration time, the Government may accept a quote (or part of a quote), whether or not there are negotiations after its receipt, unless a written notice of withdrawal is received before award.
One or more of the items under this acquisition is subject to the World Trade Organization Government Procurement Agreement and Free Trade Agreements.
It is the quoter’s responsibility to monitor the Government Point of Entry (GPE) (sam.gov) for information relevant to this solicitation, e.g., questions and answers, amendments, etc. An official authorized to bind the quoter must sign the terms and conditions of the quote. quoters that fail to complete the required representations and certifications, or reject the terms and conditions of the solicitation may be excluded from consideration.

The Government will consider for award only offers of U.S.-made or designated country end products unless the Contracting Officer determines that there are no offers for such products or that the offers for those products are insufficient to fulfill the requirements of this solicitation.
