About this file
This Sources Sought Notice is issued by the Department of Veterans Affairs (VA) Great Lakes Acquisition Center (GLAC) for single-nuclei purification, RNA sequencing, and data analysis services at the Jesse Brown VA Medical Center in Chicago, IL. The contract is a sole source, firm fixed-price procurement anticipated to last 6 months, with no option terms, under NAICS Code 541714. The project involves processing 8 samples using 10x Genomics platform with v4 chemistry, targeting 5,000 nuclei per sample, and includes comprehensive data analysis tasks such as nuclei isolation, library preparation, sequencing (approximately 200 million read pairs per sample), and advanced analytical processes like dimensionality reduction, cell clustering, and differential expression analysis.
Interested contractors must submit capability statements electronically to Michael Hrdi (michael.hrdi@va.gov) by 10:00 AM Central Time on 7/17/2025, providing detailed evidence of their ability to meet the technical requirements. The VA will evaluate submissions, with the final determination of proceeding with competitive procurement or sole source negotiation solely at the government's discretion. The principal investigator will ultimately assess any received proposals, given the highly specialized scientific nature of the research. The sources sought notice emphasizes that this is not a formal solicitation, and a Request for Proposal (RFP) may be issued at a later date.

DESCRIPTION
The Department of Veterans Affairs (VA):
Under Statutory Authority 41 U.S.C. 1901 the VA Great Lakes Acquisition Center (GLAC) intends to negotiate a sole source contract for the service of Single-nuclei purification, sequencing of RNA library using 10x Genomics platform, v4 chemistry (target 5,000 nuclei per sample), and data analysis.. The service is requested for Research Services at the Jesse Brown VA Medical Center in Chicago, IL.
This acquisition is conducted under the authority of 41 U.S.C. 1901 Simplified acquisition procedures as implemented in FAR 13.106-3(b)(3)(i), only one responsible source and no other supplies or services will satisfy agency requirements. This will be a firm fixed price contract for an estimated 6-month contract with no option terms. The NAICS Code is 541714.
The sources sought is an intent to obtain information. The statement of work is included. No solicitation documents are available and telephone requests will not be honored. However, if a firm believes it can meet the requirements it must furnish information about its products and services, as well as references from other customers who are using these products and service to the Contracting Officer by 10:00AM (CST - WISCONSIN) 7/17/2025. Supporting evidence must be furnished in sufficient detail to demonstrate the ability to comply with the above requirements. Responses received will be evaluated; however, a determination by the Government not to compete the proposed procurement based upon responses to this notice is solely within the discretion of the Government. If no responses are received, the Contracting Officer will proceed with the sole source negotiation with the contractor selected by the principal investigator of the research protocol. Any proposals received will be evaluated by the principal investigator as the expert and due to the highly scientific nature of the research. The contractor will have a direct impact on the result of the research therefore the principal investigator is the only qualified individual able to determine the ability of the contractor.
Capability statements shall be submitted by email only as a MS Word or Adobe PDF attachment to Michael Hrdi at michael.hrdi@va.gov . TELEPHONIC INQUIRIES WILL NOT BE ACCEPTED.
*If your submission exceeds 8 MB, send in multiple e-mails. Utilizing a read-receipt is advised.
NOTE: This advertisement is not a solicitation for offers, nor is it a request for proposals. A Request for Proposal (RFP) may be issued by the Department of Veterans Affairs at a later date.
STATEMENT OF WORK
1. Contract Title. Single-nuclei purification, sequencing of RNA library using 10x Genomics platform, v4 chemistry (target 5,000 nuclei per sample), and data analysis.
2. Background. Single-nuclei RNA sequencing services, including expertise in nuclei purification from various tissue, access to their library using 10x Genomics platform, and v4 chemistry (target 5,000 nuclei per sample). Their library QC and sequencing contains ~200 million read pairs of PE150 per sample and ~40,000 read pairs per nucleus. The services provided are quick turnaround, reliable, competitively priced. and found to be of high quality and efficient. Pricing must also include data support till completion of study.
3. Scope. This contract will cover the cost of Single-nuclei purification, RNA library using 10x Genomics platform, and v4 chemistry (target 5,000 nuclei per sample) Sequencing services. The project will involve 8 samples.
4. Specific Tasks.

Single-nuclei RNA library using 10x Genomics platform, v4 chemistry (target 5,000 nuclei per sample) Task 3. Library QC and sequencing: ~200 million read pairs of PE150 per sample, ~40,000 read pairs per nucleus Task 4. Data analysis includes
· Demultiplexing
· Quality control
· Sequence alignment
· Quantifying expression levels of each cell
· Secondary analysis including dimensionality reduction (PCA), cell clustering (graph-based and k-means), t-SNE, UMAP, and differential expression
· Aggregate analysis (advanced analysis) by pooling samples together for differential gene expression analysis under the given sub-populations In addition to the static differential expression tables provided for each cluster, the interactive software allows users to generate differential expression tables, heatmaps, feature plots, and violin plots dynamically for any selected groups of cells/nuclei and visualize expressions of any gene of interest across samples and treatment conditions Task 5. Electronic data transfer
5. Performance Monitoring. Report monthly via email updates on the project's progress. E-mail report sent monthly to principal investigator and COR.
6. Delivery Schedule.
Sample Number = 8 Turnaround Time: 10 working days for nuclei isolation & library
| 12 working days for sequencing |
| 5 working days for data analysis |
