About this file
This document is a Standard Form 30 Amendment of Solicitation/Modification of Contract for a U.S. Geological Survey (USGS) California Water Science Center (CAWSC) requirement. The amendment extends the response period to 08/07/2025 at 5:00am PDT and updates the Statement of Work (SOW) for DNA analysis of 100 samples from the San Francisco Bay Delta region.
The project involves collecting water samples from five monitoring stations bi-monthly between August 2025 and October 2026, with the contractor required to extract DNA from filters and conduct quantitative PCR (qPCR) analyses for specific cyanobacterial genes (mcyE, cpcB, anaC, and sxtA). Special requirements include a laboratory leader with a Ph.D. in cyanobacterial ecology, expertise in molecular methods related to cyanobacteria and cyanotoxin production, and experience working with cyanobacteria samples from California's benthic habitats. The period of performance is from 08/29/2025 to 12/31/2026, with final data deliverables due by December 31, 2026.

C. Definitions/Applicable Documents: N/A
D. Scope.
DNA preparation and qPCR
II. WORK REQUIREMENTS
A. Technical Requirements.
Chains-of-custody for all laboratory operations, and bioinformatic analyses for each sample, will be fully documented.
Samples can be received by the Contractor Monday through Friday.
Extract DNA from filter or biomass samples.
Perform qPCR for 3 cyanotoxin biosynthesis genes: microcystin (mcyE), anatoxin (anaC), saxitoxin (sxtA).
AnaC will use the primers published in: Genzoli, L., Hall, R. O., Otten, T. G., Johnson, G. S., Blaszczak, J. R., and Kann, J. (2024). Benthic cyanobacterial proliferations drive anatoxin production throughout the Klamath River watershed, California, USA. Freshwater Science
