The contract will be firm-fixed price with evaluation based on three factors of equal importance: Qualifications (personnel credentials, certifications, GLP experience required), Past Performance (minimum three HPLC analytic method validation projects within past three years with FDA or EPA experience required), and Price. The independent laboratory must be GLP-certified with a quality assurance plan and previous history performing acceptable GLP studies. Government will furnish specified antimycin A standards, samples, kits, and equipment valued at approximately $1,911. The government will award a firm-fixed price (FFP) contract resulting from this combined synopsis/solicitation to the responsible offeror whose offer, conforming to the synopsis/solicitation, will be most advantageous to the government as determined by the solicitation evaluation criteria.

STATEMENT OF WORK

I. GENERAL INFORMATION

A. Introduction

The United States Geological Survey (USGS) Upper Midwest Environmental Sciences

Center (UMESC) is seeking services to validate the antimycin A (ANT) high-performance liquid chromatography (HPLC) method. The analytical method was recently developed on the HPLC for a cost-effective widely available instrument method to quantify ANT in multiple environmental matrices (e.g., pesticide formulations). The current interlaboratory good laboratory practice (GLP) validation sought after, by a laboratory with previous experience performing GLP studies, focuses on verifying the ANT sample processing per matrix and corresponding instrumentation methodology for quantifying ANT.

B. Background

ANT is a fermentation product of Streptomyces sp. that has been used extensively as a pesticide for fish removals and fisheries management. The pesticide is a non-selective toxicant that affects cellular respiration and produces a lethal effect in fish at parts per billion concentrations. It is an ideal candidate for a pesticide targeting nuisance fish because it does not impart a detectable taste or odor and is rapidly degraded via hydrolysis and sunlight.

The quantification of total ANT is performed on a HPLC using a diode array detector

(DAD). ANT samples contain a mixture of multiple homologs. The four most abundant homologs including antimycin A1, antimycin A2, antimycin A3, and antimycin A4, in which all four homologs are quantified per sample and reported as total ANT concentration.

Going forward total ANT will be referred to as ANT(A1-A4).

D. Scope

Scope of Work:

The independent laboratory will prepare a protocol and conduct a GLP compliant analytical method validation study for a HPLC-DAD instrument method (i.e., standard calibration curve) and demonstrate the method’s ability to quantify antimycin A in multiple matrices.

The scope of work is delineated into two parts creation of an ANT(A1-A4) standard calibration curve.

1) Part 1: First, a single analyst will validate the HPLC-DAD instrument method

(following GLP study protocol) and creation of four separate antimycin A homolog (A1, A2, A3, and A4) standard calibration curve using standards prepared three separate times.

2) Option (Part 2): When part one is successful, the laboratory will perform method validation and apply the above HPLC-DAD method to quantify antimycin A homologs

(and total antimycin ANT(A1-A4)) in four different sample types following a USGS coordinated study protocol under GLP. Sample processing methods per sample type to be validated include end of fermentation brine, extracted/purified ANT, liquid ANT formulation (product), and ANT in water. USGS will provide specific sample materials, processing protocols (e.g., cleanup steps and/or concentration steps), SPE cartridges and clean up kits for each sample type.

The independent laboratory will provide a report for all validations and quantifications of ANT(A1-A4).

II. WORK REQUIREMENTS

A. Technical Requirements

Below is a list of specific reagent(s) and apparatuses necessary for the analytical method.

Reagents:

• Antimycin A from Streptomyces sp. (CAS # 1397-94-0), reagent grade or highest purity attainable.

• Ultrapure Water. 18.2 megohms-cm, from a deionization system, E-Pure

Model D4641, Barnstead (Dubuque, IA) or equivalent.

• Methanol (CAS # 67-56-1), HPLC grade.

• Acetonitrile (CAS # 75-05-8), LC-MS grade.

• Formic acid (CAS # 64-18-6), LC-MS grade.

• Anhydrous sodium sulfate (CAS # 7757-82-6), 99.0% purity, Baker

Analyzed, or equivalent.

• Ammonium acetate, ACS grade

• Acetone, HPLC grade or better

Apparatus: An HPLC system described below:

• Agilent 1290 Quaternary Pump (G7104A), 1290 Multi Column Temperature control

(G7116B), 1290 Multisampler (G7167B), 1290 Variable wavelength detector

(G1314E), QQQ LC/MS (G6460A) and Agilent Mass Hunter software for system control or equivalent LC-MS system.

• LC column, Phenomenex Kinetex 2.6 µm particle size, XB-C18 100Å, 50 × 2.10 mm or equivalent.

Laboratory equipped with the following, but not limited to:

• 12 to 24-port extraction manifold system for SPE, including vacuum pump

• LC vial caps and septa, matching cap with Teflon®/silicon septa; Waters LC-MS vial caps or equivalent product

• Minus 20 degree Celsius freezer for analytical standard and sample storage

• Powered Air Purifying Respirator (PAPR)

• Balance capable of measuring ±0.02 mg

The independent laboratory will prepare a protocol and conduct a GLP analytical method validation study for a standard operating procedure to quantify ANTA(A1-A4) in four different matrices and prepare a report. The validation should include evaluation of the following characteristics of the method performance: selectivity, instrument calibration, bias, precision, accuracy, quantitation limits and range, and detection limits.

First, the evaluation should include three individual standard curves using standards prepared three separate times and evaluation of sample processing and corresponding measurement of ANT-A(A1-A4) in all four matrices.

The analytical instrument used for the validation must be equivalent in sensitivity (e.g., response) to a HPLC using a DAD. The HPLC-DAD operating conditions for acquisition of ANT-A(A1-A4) will be provided to the independent laboratory upon award of the service contract. Each ANT homolog calibration curve should consist of at least five points ranging from approximately 35 - 1900 µg/L.

The final report must contain performance characteristics, including results compared against the following acceptance criteria: standard calibration curve for each individual homolog (A1-A4) should have an R2 ≥0.995 and contain minimum five concentration levels, accuracy of each standard ± 20% theoretical concentration, retention times variability ± 2%, k’ between 1-10, QC standard check within ± 20% of theoretical concentration, instrument precision of replicate injections ± 20% percent relative standard deviation (%RSD), and technician precision (n=3 samples %RSD ≤ 20%).

52.217-5 EVALUATION OF OPTIONS (JUL 1990) (DEVIATION MAR 2026)

Except when it is determined in accordance with FAR 17.202(b) not to be in the Government’s best interests, the Government will evaluate offers for award purposes by adding the total price for all options to the total price for the basic requirement. Evaluation of options will not obligate the Government to exercise the option(s).

52.217-8 OPTION TO EXTEND SERVICES (NOV 1999)

The Government may require continued performance of any services within the limits and at the rates specified in the contract. These rates may be adjusted only as a result of revisions to prevailing labor rates provided by the Secretary of Labor. The option provision may be exercised more than once, but the total extension of performance hereunder shall not exceed 6 months. The Contracting Officer may exercise the option by written notice to the Contractor within 30 days of before the contract expires.

52.217-9 OPTION TO EXTEND THE TERM OF THE CONTRACT (MAR 2000)

(a) The Government may extend the term of this contract by written notice to the Contractor within 30 days; provided that the Government gives the Contractor a preliminary written notice of its intent to extend at least 30 days before the contract expires. The preliminary notice does not commit the Government to an extension.

(b) If the Government exercises this option, the extended contract shall be considered to include this option clause.

(c) The total duration of this contract, including the exercise of any options under this clause, shall not exceed 2 years.

52.000S-5079 TECHNICAL LIAISON - TECHNICAL DIRECTION (OCT 1997)

a) The performance required herein shall be subject to the technical direction of the Technical Liaison (TL) as identified below. As used herein, "technical direction" is defined as direction to the contractor that fills in details, suggests possible lines of approach, or otherwise supplements the scope of the work set forth herein and shall not constitute a new assignment, and does not supersede or modify any article or clause of this contract.

(b) The Technical Liaison is not authorized to perform, formally or informally, any of the following actions:

(1) Promise, award, agree to award, or execute any contract, contract modification, or notice of intent that changes or may change this contract;

(2) Waive or agree to modification of the delivery schedule;

(3) Make any final decision on any contract matter subject to the Disputes Clause;

(4) Terminate, for any reason, the contractor's right to proceed;

GS 1131 UNILATERAL DEOBLIGATION OF UNEXPENDED FUNDS

The contractor shall submit all invoices under the award no later than 90 calendar days after the period of performance has expired, unless a request for extension has been submitted to the Contracting Officer.

After 120 days has passed since the expiration of the performance period, the government reserves the right to issue a unilateral modification deobligating any unexpended funds, and to initiate closeout procedures.

GS1396 RELEASE OF INFORMATION (JUL 2001)

(a) Disclosure of information gained as a result of work performed under this contract shall be accomplished according to U.S. Geological Survey procedures or through USGS channels. As used in this clause, the term "information" includes raw data, data derivative there from, and analysis or interpretations thereof, regardless of form. The term includes data developed or acquired by the contractor during performance of this contract, including analysis of samples provided by the Government and samples or recordings made by the contractor during contract performance.

(b) The contractor hereby agrees not to disclose such information to the public or to unauthorized parties without the prior written approval of the Contracting Officer. This restriction does not apply to releases of information to subcontractors (including consultants) as necessary for successful performance of the contract, provided (1) such information is of no commercial value to the subcontractor, and (2) the subcontractor agrees to be bound by the restrictions in this clause.

(c) After the contents of the contractor's final report have been made public by the Government, further releases of information may be made by the contractor, except for information subject to restrictions imposed elsewhere in this contract, if any. All releases must also comply with any additional restrictions which have been determined necessary by the Contracting Officer and provided to the contractor in writing.

(d) News releases pertaining to work under this contract shall not be made at any time without the approval and involvement of the cognizant Government Public Information Office.

ELECTRONIC INVOICING AND PAYMENT REQUIREMENTS - INVOICE PROCESSING PLATFORM (IPP) (APRIL 2013)

Payment requests must be submitted electronically through the U.S. Department of the Treasury's Internet Payment Platform System (IPP). "Payment requests" means any request for contract financing payment or invoicing payment by the Contractor. To constitute a proper invoice, the payment request must comply with the requirements identified in the applicable Prompt Payment clause included in the contract, or the clause 52.212-4 Contract Terms and Conditions-Commercial Items included in commercial item contracts.

The vendor shall upon invoicing through the internet payment platform, attach a copy of their regular vendor invoice by utilizing the attachments feature on the lower right hand corner of the invoicing screen to substantiate requests for payment. The vendor invoice shall be readable in Microsoft Excel, Adobe Acrobat (.pdf) or Microsoft Word.

FAR 32.905(b) outlines a Proper Invoice to include:

1. Name & address of the Contractor.

2. Invoice date & number.

3. Contract number, contract line item number and, if applicable, the order number.

4. Description, quantity, unit of measure, unit price and extended price of the items delivered.

9. Taxpayer Identification Number (TIN). The Contractor shall include its TIN on the invoice only if required elsewhere in this contract.

10. Any other information or documentation required by the contract.

The Contractor must use the IPP website to register access and use IPP for submitting requests for payment. (a) Definitions. As used in this provision-

Economically disadvantaged women-owned small business (EDWOSB) concern means a small business concern that is at least 51 percent directly and unconditionally owned by, and the management and daily business operations of which are controlled by, one or more women who are citizens of the United States and who are economically disadvantaged in accordance with 13 CFR part 127, and the concern is certified by SBA or an approved third-party certifier in accordance with 13 CFR 127.300. Service-disabled veteran-owned small business concern-

52.212-2 Evaluation - Commercial Items. (NOV 2021)

(a) The Government will award a contract from this solicitation to the responsible offeror whose offer conforming to this solicitation will be most advantageous to the government price and other factors considered. Factors 1 and 2 are of equal value and are significantly more important than cost. The following factors shall be used to evaluate offerors:

Factor 1 – Qualifications: Contractor shall provide a listing of personnel performing the work, their laboratory credentials, certifications, awards and accreditation for validation following good laboratory practices. All work must be performed at a certified laboratory, by qualified technicians. Listings should include education, work experience and number of years employed in this capacity. Previous experience working with good laboratory practice (GLP) is essential.

Factor 2 – Past Performance: Contractor shall provide references for a minimum of three (3) projects within the past 3 years related to performing HPLC analytic method validation services. Prior experience with Food and Drug Administration or U.S. Environmental Protection Agency analytic method validation is a requirement of this category. Each reference shall include: the company or agency name, address, contract or order number, point(s) of contract phone number and email address(es). References provided may be contacted by the Government. Their statements will be used in the evaluation of the offeror’s quotation.

In addition, any information obtained by the Government from references outside of the offeror’s quotation may also be used, in a similar manner, in the technical evaluation of the offeror’s quotation to the extent appropriate. Past performance information provided by references, from Government records, or from other than the sources identified by the offeror may be used in the evaluation under any or all applicable evaluation factors. Contractor Performance Assessment Reporting System (CPARS) reports and Federal Awardee Performance and Integrity Information System (FAPIIS) records will be used as a source of past performance information in the source selection process, in addition to any other past performance information required under this solicitation. All past performance information will be used for both the responsibility determination and the best value decision.

(b) A written notice of award or acceptance of an offer, mailed or otherwise furnished to the successful offeror within the time for acceptance specified in the offer, shall result in a binding contract without further action by either party. Before the offer’s specified expiration time, the Government may accept an offer (or part of an offer), whether or not there are negotiations after its receipt, unless a written notice of withdrawal is received before award.

Award The award of this requirement shall be made using only the procedures found in Part 12 of the Federal Acquisition Regulations (FAR). Award shall be made to the vendor whose quotation, conforming to this RFQ, is determined to be the most advantageous to the government, cost or price and the other non-cost or non-price factors listed above. In determining which quotation offers the greatest value or advantage to the Government, overall price or cost to the Government will be comparable in importance to overall technical merit. Between substantially equal technical quotations, the proposed evaluated price or cost will become the major factor in selection for award. Between acceptable quotations with significant difference in technical rating, a determination will be made as to whether the additional technical merit or benefits reflected by a higher priced quotation warrants payment of the additional price or cost.

A single award shall be made to the vendor whose proposal, conforming to this Solicitation, it determined to be most advantageous to the government, cost or price and other non-cost or non-price factors listed above. In determining which proposal offers the greatest value or advantage to the Government, overall past performance and qualifications will be significantly more important than evaluated price or cost to the Government. Price or cost will become the determining factor between proposals judged to be essentially equal in technical merit. While the Government anticipates award based primarily on technical superiority, no award will be made based on superior technical capability when the proposed cost or price is considered unreasonable or when the additional cost or price is not justified by the advantages of an award based on technical superiority.

The offeror is cautioned to study the Evaluation Factors stated above along with their relative importance.

Your proposal package shall address each of the factors listed above since these are the standards against which your proposal will be evaluated. It is the offeror's responsibility to include the appropriate documentation in its proposal package to allow a complete technical and price evaluation. Failure to submit the appropriate documentation may result in the rejection of the offeror's proposal.

All technical proposals shall be separate from pricing proposals. No pricing shall be contained in the technical proposal. Offerors are required to list discounts, if any, as part of their pricing proposal, but separate from the original unit price.
