Questions and Answers 09 through 10
09. Please specific the rate and other specifics for quality controls for pricing purposes.
a. How many LFBs (lab fortified blanks) and LFSMs (lab fortified sample matrix) are required per batch?
ANSWER. Under the awarded Indefinite Delivery contract, the task orders and micropurchase orders will have various quantities per "batch"; thus, each offeror shall price its minimum "batch" quantity and discount price points for larger "batches" using the Attachment 1 Price Schedule template.
b. How many samples are there per batch?
ANSWER. Under the awarded Indefinite Delivery contract, the task orders and micropurchase orders will have various quantities per "batch"; thus, each offeror shall price its minimum "batch" quantity and discount price points for larger "batches" using the Attachment 1 Price Schedule template.
c. ELISA samples are already analyzed in duplicate per manufacturer instructions, or do you mean that a second extract is to be processed and analyzed for 100% samples via ELISA and 20% for LC-MS/MS. Or do you intend for the same extract (same vial) to be analyzed in duplicate 100% ELISA and 20% LC-MS/MS?
ANSWER. ELISAs will be analyzed in duplicate per manufacturer's instructions. 20% of LCMSMS samples will be analyzed in duplicate per project requirements.
d. Since the SPATT is not collected in duplicate for QAQC purposes, do you intend for the LFSMs to be prepared in the extracts from SPATT? Do you want all 3 extracts to be spiked from a single SPATT, or a single extract (first, second, or third extract)? Please clarify how spiking (LFSMs) should be conducted.
ANSWER. Lab fortified sample matrix is environmental sample spiked with concentration of known standard. "Environmental sample" can be water or SPATT extract. In each ELISA run, a certain percentage of samples should be spiked with a known concentration of the toxin being analyzed. These should be selected at random. of water and SPATT extract samples using QA/QC samples - including LFSM - as outlined in EPA 546. Contractor will provide USGS with specific spiking procedures with their bid.
10. Matrix effects caused by interferences from SPATT extracts are not addressed in the ELISA methods and low MRLs (kit based DLs) run the risk of false positive data. Since we have not seen previous data…. would the USGS accept interpretations of data that reflect the inherent limitations of ELISA based data from SPATT resin extracts in reporting?
ANSWER. Yes.
