Algal Toxin and Related Analysis: SPATT Extraction and Analysis and Whole Water Toxin Analysis

GENERAL INFORMATION

A. INTRODUCTION

The Department of the Interior (DOI) U.S. Geological Survey (USGS) Upper Midwest Water Science Center’s (UMid-WSC) has a requirement for a Indefinite Delivery Requirements (IDR) Contract for Solid Phase Adsorption Tracking Tool (SPATT) extraction and analysis services.

The USGS Upper Midwest Water Science Center’s (UMid-WSC) works in collaboration with several other agencies to document cyanotoxins in Minnesota, Wisconsin, Michigan, in addition to several surrounding states and at National Parks across the Nation. The UMid-WSC needs an experienced provider of Solid Phase Adsorption Tracking Tool (SPATT) extraction and analysis of numerous algal toxins by both tandem liquid chromatography mass spectrometry (LCMSMS) and enzyme-linked immunosorbent assay (ELISA).

B. PROJECT BACKGROUND

The UMid-WSC Harmful Algal Bloom and Integrated Ecosystem Services Teams are involved in several projects dealing with cyanotoxins. These projects include the documentation and prediction of harmful algal blooms (HABs), the diel variability of neurotoxins, relation of cyanotoxins to water levels, and research related to rapid-response strategies for identifying cyanotoxic HABs. Several of these research studies incorporate SPATT technologies in order to better understand the relations between cyanotoxin occurrence and environmental factors.

Historically, the requirement for these analysis services have been:

So far, no analyses have been ordered in FY 2021 (from 10/01/2020 to 05/31/2021).

The USGS estimates, but by no means guarantees, the following analyses over the 5.5-year period of performance, if all options are exercised. These quantities are strictly estimated for USGS budgeting purposes only. These estimates are not to be considered as guaranteed work. The Government will order work only when there is a bonafide need and funding available.

C. SCOPE

Services, non-personal, to provide all equipment, supervision, labor, travel, and supplies and materials (unless otherwise provided herein) necessary to provide SPATT extraction and analysis, LCMSMS analysis and ELISA analysis. The purpose of this requisition is to obtain algal toxin extraction and analysis from SPATT) samplers and water samples by LCMSMS and ELISA and evaluate these sampling methods.

PROJECT SPECIFICATIONS AND ASSOCIATED COSTS

The UMid-WSC staff will be responsible for collecting and processing all samples and submitting all administrative paperwork and samples to the contractor (laboratory). The UMid-WSC has several ongoing projects that require SPATT extraction and analysis and toxin analysis (LCMSMS and ELISA). These projects vary in regard to sample size, sample number, and type of sensors required for the buoys. Several of these projects are multi-year projects and to ensure consistency and data quality on these projects, analyses shall be performed by the same contractor.

D. TECHNICAL REQUIREMENTS

When ordered by the Government:

The contractor shall receive samples on Federal workdays.

The contractor shall document the chains-of-custody for all laboratory operations, and analyses for each sample.

The contractor shall extract algal toxins from SPATT samples using methods described by Howard et al.

(2018).

The contractor shall analyze the extracted samples for algal toxins using LCMSMS by previously published methods (described below). Once the analyses are run, the three extract values shall be added together to get a toxin value that is normalized to the dry weight of the resin, grams of resin, and length of deployment.

The contractor shall analyze the SPATT and water samples for Microcystins, Nodularin, Cyanopeptolins, and Anabaenapetins using methods described by Bartlett et al. (2018). Briefly, the cyanotoxins will be measured in 20 μL injections using LCMSMS with electrospray ionization.

The contractor shall separate analytes on a C18 column using a Shimadzu Prominence HPLC and quantified on a Sciex 4000 QTRAP with electrospray ionization using a scheduled multiple reaction monitoring method.

The contractor shall analyze samples for Cylindrospermopsin, Homoanatoxin-a, Anatoxin-a, Tetrodotoxin, Saxitoxin, Decarbamoyl Gonyautoxins, Gonyautoxins, Decarbamoyl Neosaxitoxin, Decarbamoyl Saxitoxin, and Neosaxitoxin using methods described by Beversdorf et al. (2017).

The contractor shall measure cyanotoxins in 15 μL injections using LCMSMS with electrospray ionization and hydrophilic interaction chromatography.

The contractor shall extract Cyanotoxins (microcystin, saxitoxin, and cylindrospermopsin) from the SPATT samplers and shall analyze by three separate enzyme-linked immunosorbent assays (ELISA) acquired from Abraxis LLC / Eurofins (Warminster, PA) for microcystins/nodularins, saxitoxins, and cylindrospermopsins, (Graham et al., 2010). The minimum reporting levels (MRL) are 0.05 μg/L as cylindrospermopsin equivalents, 0.10 µg/L as Microcystin-LR equivalents, and 0.02 μg/L as saxitoxin equivalents.

The contractor shall dilute samples exceeding the highest calibration standard. The contractor shall notify USGS regarding any samples that do not pass quality control checks.

GENERAL REQUIRMENTS

The contractor shall:

1. Deliver data no later than 60 days following receipt of samples. Data shall be accompanied by a spreadsheet listing sample identifiers.

2. Provide a written document describing the methods used for all processes including all details on reagents, kits, and equipment used. This document shall be provided with data no later than 60 days following receipt of samples.

3. The contractor shall provide information on technology, technical requirements, and answers to questions regarding services upon request.

USGS QUALITY ASSURANCE SURVEILLENCE PLAN

The USGS Technical Liaison (TL) will assure that the contractor delivers the quality products and services described. The TL will monitor all aspects of the services performed by the contractor. Substandard Services, services that fall short of the described standards, shall be re-done by the contractor at no additional cost to the Government within 30 days of identified issue.

Contract Type- REQUIREMENTS CONTRACT This is an Indefinite Delivery, Requirements Contract.

The Department of the Interior (DOI) U.S. Geological Survey (USGS) Upper Midwest Water Science Center (UMid-WSC) 8505 Research Way, Middleton, Wisconsin 53562 will order required services, within the scope of this contract, at the Price Schedule prices, via unilateral modifications to this contract

Except as this contract otherwise provides, the Government shall order from the Contractor all the supplies or services specified in the Schedule that are required to be purchased by the Government activity or activities specified in the Schedule.

The Government is not required to purchase from the Contractor requirements in excess of any limit on total orders under this contract.

If the Government urgently requires delivery of any quantity of an item before the earliest date that delivery may be specified under this contract, and if the Contractor will not accept an order providing for the accelerated delivery, the Government may acquire the urgently required goods or services from another source.

Any order issued during the effective period of this contract and not completed within that period shall be completed by the Contractor within the time specified in the order. The contract shall govern the Contractor’s and Government’s rights and obligations with respect to that order to the same extent as if the order were completed during the contract’s effective period; provided, that the Contractor shall not be required to make any deliveries under this contract after 90 days after contract expires.
