WORK STATEMENT
I. GENERAL INFORMATION
A. Introduction.
The Department of the Interior (DOI) U.S. Geological Survey (USGS) Upper Midwest Water Science Center (UMid WSC), 2280 Woodale Drive, Mounds View MN 55112 has a requirement for multi-endpoint gene regulatory assays to evaluate cellular responses to organic chemicals extracted from groundwater samples in petroleum hydrocarbon contaminant plumes.
B. Background.
In 2016, our research group published a journal article showing that two crude oil contaminant plumes contain complex mixtures of partial transformation products from biodegradation of the crude oil (Bekins and others, 2016). The total concentration of these compounds is over two to three times higher than that represented by the standard analysis required by regulators. Current knowledge indicates that there are thousands of different compounds present and only a fraction of them have been identified. Thus, it is not possible to assess the toxicity by analyzing the concentrations of individual compounds. Working with a toxicologist, we have determined that the best approach is to measure biological responses to the entire collection of compounds in the water.
The USGS is presently conducting a series of coordinated studies to understand the nature and toxicity of partial transformation products associated with petroleum hydrocarbon sources.
Some of the studies are taking place at the USGS Bemidji Crude Oil Research site. Others are to be carried out on one or more groundwater plumes at refined fuel spill sites in Minnesota and elsewhere. The data from these studies will be used to determine how much additional research is needed to determine if drinking water and the environment will be impacted by these compounds.
C. Definitions/Applicable Documents
Environmental sample: water sample collected by the USGS from an environment of interest.
Sample may be taken from a groundwater or surface-water source. Samples may include multiple dilutions or technical replicates; samples are the primary unit of measure being tested for each Contract Line Item (CLIN).
Assay_Endpoints_Table1_2.xlsx: contains 2 tables that list the assay endpoints for which we are requesting analysis. Table 1 lists 46 transcription factor pathways and table 2 lists 48 human nuclear receptors
Bekins, Barbara A., Isabelle M. Cozzarelli, Melinda L. Erickson, Ross A. Steenson, and Kevin A. Thorn. "Crude oil metabolites in groundwater at two spill sites." Groundwater 54,
no. 5 (2016): 681-691.
D. Scope The USGS requires a contractor to provide services, non-personal, necessary for completing multi-endpoint gene regulatory assays with live cells. The contractors shall use technologies for profiling transcription factor activity in cell lines and cell culture conditions appropriate for toxicity testing when performing these services. Each assay shall address multiple transcriptional factor targets within a single well.
The contractor shall provide all equipment, labor, travel, and materials (unless otherwise provided herein) necessary to complete the tasks outlined in the Work Statement (PWS). This includes general contract management and administrative functions such as meetings and teleconferences with USGS to review data, issues, and performance.
II. WORK REQUIREMENTS
USGS personnel will collect environmental water samples. Researchers collaborating with USGS will extract the organic compounds from the water samples. These extracts will be frozen and then shipped overnight to the contractor for analysis.
A. Technical Requirements.
The contractor shall receive shipped environmental sample extracts, maintain Quality Control over them, analyze them, and provide resulting data and metadata to the USGS. The contractor shall elute and reconstitute extract samples for use in the assays. Environmental samples shall be analyzed with several multi-endpoint assay types (listed below). Each environmental sample will be analyzed, at a minimum, in duplicate for each assay type. Contractor shall be capable of diluting or concentrating environmental samples such that each environmental sample may be analyzed at multiple concentrations. The contractor shall minimize intra-assay variation by simultaneously assessing multiple endpoints in a single well of cells.
At a minimum,
1. The contractor shall be capable of simultaneously analyzing environmental samples for effects on 46 transcriptional pathways in human cell lines (listed in table 1).
2. The contractor shall be capable of simultaneously analyzing environmental samples for effects on 48 human hormone nuclear receptors in human cell lines (listed in table 2).
Additionally,
3. The contractor shall provide a price list and target endpoint list for other multi-endpoint gene regulatory transcription factor assays that the contractor is capable of performing.
B. Deliverables.
The contractor shall provide data to the USGS in electronic formats via email or other acceptable forms of delivery.
1. Metadata, environmental sample data, and relevant quality assurance and quality control data for work product described in II.A.1., identified by name and quantity.
2. Metadata, environmental sample data, and relevant quality assurance and quality control data for work product described in II.A.2., identified by name and quantity.
3. Metadata, environmental sample data, and relevant quality assurance and quality control data for work product described in II.A.3., identified by name and quantity.
4. Delivery dates for work/product to be completed and point of delivery. Contract lab analytical results for environmental samples will be delivered not later than 90 days after samples are received at the contractor’s facility
III. SUPPORTING INFORMATION
A. Place of Performance.
Contractor’s designated laboratory
B. Periods of Performance: This blanket purchase agreement will be valid for 5 years after the execution of the agreement.
C. Government Furnished Property (GFP) None
QUALITY ASSURANCE SURVEILLANCE PLAN
The Contractor shall be responsible for Quality Control. The Contractor shall provide a Quality Control Plan with their offer/proposal/quote; and shall adhere to the Government accepted QCP throughout performance of this contract. Samples will be run in duplicate. All Standard Operating Procedures (SOPs) used under this agreement shall include adequate Quality Control (QC) to ensure the data are within acceptable error limits and are of known, acceptable, and verifiable quality. At a minimum, the Contractor shall include quantitative criteria for the decision to pass or fail test data for each assay on a per plate basis.
The pass/fail criteria will typically be based on positive and negative controls run on the same plate as samples and on results that indicate patterns significantly different from normal. Negative controls reflect responses to the solvent alone. In this case the solvent is expected to be DMSO. Positive controls reflect responses of chemical known to cause upregulation in the assays. Ideally a hydrocarbon such as benzene or naphthalene should be included in the positive controls. The Contractor shall repeat the assay(s) for those that do not meet the appropriate QC standard at no cost to the Government.
The Government will be responsible for Quality Assurance. The USGS Technical Liaison (TL) or Technical Contracting Officer Representative (COR) will review data from the Contractor’s facility to ensure that the Contractor is meeting its stated quality standards. Additionally, the TL/COR may opt to conduct external performance audits with independent quality assurance samples that verify the contractor’s analytical performance. Quality assurance samples could include blank samples, replicate samples, or known standards. The USGS TL/COR will notify the contractor of any substandard performance. The contractor will reperform all required services for substandard results within 30 days after notified of substandard performance.
OFFERS SHALL INCLUDE A PRICE SCHEDULE.
The following table is a sample of an acceptable pricing schedule. Offerors may use their own price schedule format. Offerors may use current pricing that may be increased within price reasonableness with a 30 day notice.

(a) The Contracting Officer will screen proposals to identify and eliminate any proposal that are so incomplete as to preclude a meaningful evaluation of technical and past performance issues.

References provided may be contacted by the Government. Their statements will be used in the evaluation of the seller’s bid package. In addition, any information obtained by the Government from references outside of the seller’s bid package may also be used, in a similar manner, in the non-price evaluation of the seller’s bid to the extent appropriate. Past performance information provided by references, from Government records, or from other than the sources identified by the seller may be used in the evaluation under any or all applicable evaluation factors. In addition to the foregoing, PPIRS.gov will be used as a source of past performance information in the source selection process, in addition to any other past performance information required under this solicitation. In determining which offer offers the greatest value or advantage to the Government, overall non-price merit will be considered more important than price or cost. The degree of importance of price or cost as an evaluation factor will increase with the degree of equality in the non-price merits of the offers. Between acceptable offers with a significant difference in non-price merit, a determination will be made as to whether the additional non-price merit or benefits reflected by a higher priced offer warrants payment of the additional price or cost.
