About this file

This document is a Standard Form 30 Amendment of Solicitation/Modification of Contract (Amendment 0001, effective 07/14/2026) issued by the U.S. Fish and Wildlife Service's Indiana Ecological Services Field Office (FWS SAT Team 2, Falls Church VA 22041) for solicitation 140FS226Q0141.

The amendment provides government-furnished information regarding a bat genomic assessment contract (FWS PPR #2117692) with a performance period of 10/01/2026 to 12/31/2027. The government requires genomic sequencing services for approximately 200-400 myotis (MYSO) wing punch samples with sequencing depth greater than 15-20x coverage per individual. Sample portions are currently stored at the University of Illinois (Dr. Mark Davis) and the Indiana Department of Natural Resources in Bloomington. Previously processed samples prepared using ddRADseq methodology (Sacks protocol with MspI and PstI restriction enzymes, 370-720 bp size selection, Illumina NovaSeq 6000 platform, 150 bp paired-end reads) will be shared with the awarded contractor. Offerors must propose sequencing approaches compatible with the existing dataset, either using the same ddRADseq methodology or alternative approaches such as Whole Genome Sequencing, provided resulting data maps to the same reference genome. Low-coverage sequencing will not be accepted. Quotations must clearly identify all costs associated with meeting specified data quality requirements. The deadline for acknowledging receipt of this amendment is the hour and date specified in the original solicitation, with failure to acknowledge resulting in potential offer rejection.

IN - INDIANA ES FIELD OFFICE - Bat Genomic

Assessment, 0001

FWS PPR #2117692

The Government is providing the following additional information to assist offerors in preparing quotations.

Sample Storage

The U.S. Fish and Wildlife Service's Indiana

Ecological Services Field Office (INFO) does not have an on-site -80°C freezer. As a result:

-A portion of the MYSO wing punch samples are currently being stored by Dr. Mark Davis at the

University of Illinois.

-Additional MYSO samples are being stored by the Indiana Department of Natural Resources in

Bloomington, Indiana.

Previously Processed Samples

-Over the past year, the Davis Laboratory (Dr.

Mark Davis and Jordyn Chase) processed a subset of the MYSO samples, at no cost to the

Government, while utilizing available capacity during sequencing for a separate research project.

- The Davis Laboratory understands that these data will be shared with the contractor ultimately awarded this requirement.

The Government is currently coordinating with the Davis Laboratory to determine:

-The exact number of samples that have already been processed; and

-Which of those samples will be included in the final genomic analysis under this requirement.

**This information will be provided once finalized.

Compatibility Requirements

Because a portion of the samples has already been processed, offerors must ensure that their proposed sequencing approach is compatible with the existing dataset.

Offerors may propose using the same library preparation and sequencing methodology employed

by the Davis Laboratory (ddRADseq), or they may propose an alternative approach. Regardless of the methodology proposed, the quotation must satisfy all requirements identified in the

Statement of Work, including the required sequencing depth and data quality (e.g., sequencing approximately 200-400 individuals at greater than 15-20x coverage per individual).

Low-coverage sequencing will not be accepted.

Quotations should clearly identify all costs associated with meeting these data quality requirements.

The previously processed samples were prepared using the following methodology:

-Library preparation: ddRADseq using the Sacks protocol

-Restriction enzymes: MspI and PstI

-Size selection: 370-720 bp (final library size with adapters; approximately 370-680 bp prior to adapter addition)

-Sequencing platform: Illumina NovaSeq 6000

-Read configuration: 150 bp paired-end reads

-Additional details regarding the library preparation protocol can be found in the referenced dissertation (page 90 of the dissertation; page 96 of the PDF including front matter):

https://www.ideals.illinois.edu/items/126823

Please note:

- If proposing additional ddRADseq work, the library preparation protocol must be compatible with the methodology described above so that the datasets can be combined.

- Whole Genome Sequencing (WGS) may also be proposed; however, the resulting data must be compatible with the existing ddRAD dataset, including mapping to the same reference genome.
