About this file

This is a Statement of Work (SOW) for the acquisition of an automated cell counting and assessing device for the USDA Agricultural Research Service in Ames, Iowa. The device must be capable of counting and assessing the viability of hundreds of thousands of cells (5-80 microns in size) for single cell sequencing and gene expression research.

The technical requirements specify a benchtop instrument (maximum dimensions 10" x 14" x 19") that can process 8 samples simultaneously, calculate size/count/concentration/viability in under 60 seconds, and handle various sample types including cultured cells, PBMCs, and whole blood. Key features include dual fluorescence and brightfield imaging, glove-friendly touchscreen, automatic slide detection, LED illumination, and compatibility with disposable/shatterproof slides. The unit must be delivered within 60 days of award to the USDA facility in Ames, IA. Evaluation factors prioritize technical capability and deliverability over price, with one complete system to be provided.

Statement of Work Project: Acquisition of an Automated Cell Counting and Assessing Device

GENERAL INFORMATION

1.0 Scope of Work: Acquisition of an automated device that can count and assess the viability of cells for single cell sequencing for gene expression.

2.0 Background: Acquisition of an automated device that can calculate the size, count, concentration, and viability of cells used generate single cell partitions cells for transcriptomic and epigenomic research. Must be able to count and assess the viability of hundreds of thousands of cells 5-80 microns in size.

CONTRACTOR REQUIREMENTS

3.0 Technical Requirements/Tasks:

The automated cell counting device must meet the following minimum specifications:

o Must be capable of counting and assessing the viability of hundreds of thousands of cells o Must be capable of calculating the size, count, concentration, and viability of cells in less than 60 seconds o Must be compatible with sample types including cultured cell lines, PBMCs, primary cells, and whole blood o Must utilize both dual fluorescence and brightfield imaging in processing o Must have cell size flexibility for cells 5-80 microns in size o Must be capable of counting a minimum of 8 samples at a time o Must be capable of processing volumes as small as 10ul o Must allow user to customize experiment settings and save commonly used assays o Must have both slide and image-based autofocus methods o Must have bi-directional stage movement to capture maximum sample volume o Must detect sample slides automatically o Must begin counting as soon as the slide is inserted o Must illuminate samples with high power LEDs o Must include a glove-friendly touchscreen o Must include an on-screen keyboard for custom naming of files o Must be able to perform multiple replicates within a single slide o Must be compatible with disposable slides for infectious samples o Must be compatible with slides composed of shatterproof plastic materials o Must be compatible with slides that have mitigated cross-contamination risk o Must include supply and power cord o Must have at least 2 ports on the front and back of the instrument o This unit should be plug-in and run, requiring minimal installation other than to level and check functionality o System shall be benchtop style instrument with dimensions not to exceed 10” wide x 14” deep and no more than 19” tall.

o Qty: 1 system.

4.0 Deliverables/Schedule This unit should be delivered onsite within 60 days of date of award.

5.0 Evaluation Factors Technical capability, deliverability and price with technical capability and deliverability more important than price.

6.0 Technical Point of Contact: Should there be technical questions, please contact
