Statement of Work (SOW) Project: Enrichment and selection of resistance (R) genes from non-model organisms
GENERAL INFORMATION
1.0 Scope of Work:
To provide biotinylated RNA baits for targeted enrichment capture of DNA-based R genes in two non-model organisms, and target enrichment kits compatible with NGS library preparation kits from.
This is in fulfillment of an ARSX-funded project aimed to identify the total number of resistance genes in USDA GRIN core plant collections of citrus and sugar beet.
2.0 Background:
The East Lansing, MI Sugar Beet and Bean Unit conducts research on genetic diversity and genomic tool development for sugar beets and other crops. The biotinylated RNA baits are an essential component for the completion of an ARSX-funded project emphasizing germplasm diversity and maintenance. Traditional sequencing is not sufficient for resistance gene discovery, and special systems must be used. The targeted enrichment of R genes prior to sequencing is crucial, and this requires RNA-based baits to bind and select resistance genes from whole genomic DNA.
CONTRACTOR REQUIREMENTS
3.0 Technical Requirements/Tasks:
• Provide biotinylated RNA-based baits (40-60k) for resistance genes in citrus and ensure that they are fully functional.
• Provide biotinylated RNA-based baits (60-80k) for resistance genes in sugar beet and ensure that they are fully functional.
• Baits must be between 60 and 120 nucleotides in length.
• Supply enrichment kits with sufficient reactions for citrus (48 rxns) and sugar beet (16 rxns) that contain all needed specialty reagents.
4.0 Deliverables / Schedule:
All items must be delivered to 1066 Bogue Street, Room 361, East Lansing, MI 48824 by March 15, 2025.
