About this file
This document contains industry questions and government responses regarding solicitation number 12505B23Q0252 from the Department of Agriculture Agricultural Research Service Field Research Implementation and Information Delivery Midwest Area for DNA sequencing services. The government requests sequencing of PCR products from 16S and 18S primers amplified from 312 bacterial and fungal samples. The samples will be provided as genomic DNA for the contractor to complete PCR amplification and add barcodes and adapters prior to sequencing.

Industry Questions and Government Responses
RFQ 12505B23Q0252
DNA Sequencing
1. Question: Can you clarify that sequencing is requested for up to 624 PCR products (16S + 18S) taken from 312 samples?
Response: Yes, we request sequencing of 612 samples. We have tested both 16S and 18S primers on all 312 organismal samples and confirmed amplification of a single or a few with double band.
2. Question: A little more information on the PCR product to be delivered will be helpful. For example, will samples be delivered as PCR product, then needing adapters and barcodes added, or will PCR product be delivered already prepared with adapters and barcodes?
Response: The samples will be genomic DNA with The CD Genomics. They would do the amplification with our specified 16S and 18S primers and add barcodes and adapters. Below is excerpt from their email:
• Are you able to just provide the genomic DNA and we do the PCR amplification? Because even if you provide the PCR products, we still need to do another round of PCR to make the libraries. "
