About this file
This is a Questions and Answers document for RFQ# 1232SA26Q0845 addressing clarifications on technical requirements and eligibility for submission.
The Government requires a single detection platform capable of simultaneously detecting metabolites, lipids, proteins, and other small compounds within one chromatographic acquisition, eliminating the need for multiple assays or sequential acquisitions. Sample volume requirements specify a minimum of 100 microliters but no more than 250 microliters per submission, with the remaining plasma reserved for other assays. For proteomics analysis, vendors should utilize publicly available Sus scrofa (pig) sequences from standard databases; while the reference strain is primarily Landrace by Yorkshire with residual Duroc genetics, previous works have successfully employed the publicly available Sus scrofa sequences. This RFQ is currently set aside for small business vendors only. Public universities and other than small business concerns are ineligible to submit unless the solicitation is reposted as unrestricted following receipt of no quotes or no technically acceptable quotes.

Questions and Answers for RFQ# 1232SA26Q0845
Q: Definition of "simultaneous" detection: Attachment 2, Section 2.0 states that "spectral data will simultaneously detect metabolites, lipids, proteins, and other small compounds," while Section 4.0 references "one-time sample preparation." Does the Government require that all molecular classes be detected within a single chromatographic acquisition, or is a single, one-time sample preparation followed by multiplexed acquisitions with integrated cross-class data processing, statistical analysis, and pathway enrichment acceptable?
A: To reduce experimental error and quantity of sample needed, we require all molecular classes be detected on a single platform.
Q: We will need multiple assays to satisfy this requirement. Can you confirm if this is expected or not?
A: A single detection platform/ run is expected.
Q: We will require minimum 100ul for all the assays. Can you confirm if this is suitable for this project?
A: A minimum of 100 ul but no more than a maximum of 250 ul can be provided as we need the remaining plasma for other assays.
Q: The pig reference proteome is Sus scrofa (Duroc) on Uniprot database
(https://www.uniprot.org/proteomes/UP000008227). Can you confirm if this is suitable for proteomics assay?
A: The breed is primarily Landrace by Yorkshire but does have some residual Duroc from previous generations. Previous works have utilized the Sus scrofa sequences that are publicly available.
Q: Would a public university be able to submit a quotation for this project?
A: This RFQ is currently set aside for small business vendors only. As such, unless no quotes are received or no technically acceptable quotes are received and the solicitation is reposted as unrestricted a public university (i.e. an Other Than Small Business Concern) would be unable to submit a quote that could be put into consideration for an award.
