Description

Abstract
Regulation of X chromosome gene expression is essential for proper development and homeostasis in mammals, and disruptions in this process can lead to developmental disorders and cancer. Xist, a long noncoding RNA, is well-established as the key regulator of X chromosome inactivation (XCI) in females. Xist expression has never been detected in any male animal, making its presence in male peromyscus leucopus a novel and unexpected finding. My research focuses on the expression of xist in male peromyscus leucopus and its interaction with the repressive histone modifier H3K27ME3, which is associated with gene silencing. My preliminary data from RNA fish and immunofluorescence demonstrate colocalization of xist RNA with H3K27ME3, suggesting a role for xist in regulating the male X chromosome. The goal of this project is to determine how xist interacts with chromatin and contributes to gene silencing on the male X chromosome in P. Leucopus. This will be achieved through two aims: first, to map xist-associated chromatin regions and their H3K27ME3 landscape using chip-SEQ combined with single-cell sequencing and RNA fish, and second, to assess the functional consequences of xist loss through mutagenesis and RNA-SEQ. These experiments will uncover mechanisms by which xist contributes to chromatin organization and transcriptional regulation in males, addressing a significant GAP in our understanding of X chromosome biology. My research is supported by the expertise of my mentors. DR. SHA sun, my sponsor, specializes in xist biology and X chromosome regulation, and DR. Anthony long, my CO-sponsor, brings extensive knowledge of peromyscus genetics and evolution. With their guidance and a comprehensive training plan, I will develop the skills needed to successfully complete this project and contribute to our understanding of X chromosome regulation and evolution.
