(i) This is a combined synopsis/solicitation for commercial items prepared in accordance with the format in Subpart 12.6 as supplemented with additional information included in this notice. This announcement constitutes the only solicitation; proposals are being requested and a written solicitation will not be issued. (ii) The solicitation number is 75N95019Q00267 and the solicitation is issued as a request for quotation (RFQ). This acquisition is for a commercial item or service and is conducted under the authority of the Federal Acquisition Regulation (FAR) Part 13-Simplified Acquisition Procedures; FAR Subpart 13.5-Simplified Procedures for Certain Commercial Items; and FAR Part 12-Acquisition of Commercial Items, and is NOT expected to exceed the simplified acquisition teshold at $250,000. THIS IS A NON-COMPETITIVE (NOTICE OF INTENT) COMBINED SYNOPSIS SOLICITATION TO AWARD A CONTRACT OR PURCHASE ORDER WITHOUT PROVIDING FOR FULL OR OPEN COMPETITION (INCLUDING BRAND-NAME). The Office of Acquisition (OA) at the National Institute on Drug Abuse (NIDA), on behalf the TRND-Relaxin FIU project in the Therapeutic Development Branch (TDB) at the National Center for Advancing Translational Sciences (NCATS), intends to negotiate and award a purchase order without providing for full and open competition (including brand-name). This is a Brand-Name Only requirement for the HTRF Pro-collagen Type 1 (human) kit as identified herein that is manufactured by Cisbio US, Inc., located at 135 South Road, Bedford, MA 01730-2204. This acquisition is conducted as non-competitive for a commercial item or service and is conducted pursuant to FAR 13.106-1(b)(1). The rationale for the brand-name justification is pursuant to the market research conducted, specifically the sources sought notice number 75N95019Q00267 released on FedBizOpps from July 17, 2019 tough July 29, 2019 specifically identifying no other source technically capable of achieving this requirement. (iii) The solicitation document and incorporated provisions and clauses are those in effect tough Federal Acquisition Circular (FAC) Number 2019-05, with effective date August 13, 2019. (iv) The associated NAICS code is 325414- Biological Product (except Diagnostic) Manufacturing with size standard 1250 employees. A Small Business Set-Aside restriction is applicable. (v) This requirement is to obtain quantity six (6) HTRF Pro-collagen Type 1 (human) kits as identified herein that is manufactured by Cisbio US, Inc., located at 135 South Road, Bedford, MA 01730-2204. The assay reagent kits are needed to run in vitro cell based assays to support medicinal chemistry for the TRND-Relaxin FIU project in the TDB. The Dharmacon siRNA RNAi plates provide high flexibility for selecting plate layout and target genes. This library contains genes that are considered potential targets for therapeutics, which include, kinases, proteases, GPCRs, ion channels, phosphatases, and DNA repair. The siRNA designs and modifications reduce off target effects while maintaining high silencing potency for high confidence screening results. This chemistry produces clearer results providing, effective knockdown, higher specificity, greater stability and less cellular toxicity. Specifically, the following are considered essential characteristics of this requirement: Must be detectable in a sandwich assay format using 2 different specific monoclonal antibodies, one labeled with Europium Cryptate (donor) and the second with d2 (acceptor). Must allow for low volume sample size needed. Must be good for live cell, supernatant, and biochemical-based protocols. Kit must come as all-in-one ready to use kit with no necessity of additional purchase of components to make it work. Option for simple "add & read" protocol, no washing step. Sandwich immunoassay detecting human pro-collagen type 1 Must allow detection of human pro-collagen type 1 in cell supernatants. Must allow quantification of human pro-collagen type 1 in cell supernatants. Must allow functional screening of compounds using Human pro-collagen type 1 secreting cells. BACKGROUND The National Institutes of Health (NIH) is the nation's leading medical research agency and the primary Federal agency whose mission is to seek fundamental knowledge about the nature and behavior of living systems and the application of that knowledge to enhance health, lengthen life, and reduce illness and disability. The National Center for Advancing Translational Sciences (NCATS) focuses on getting more treatments to more patients more quickly. Several thousand genetic diseases affect humans, of which only about 500 have any treatment. NCATS is directly addressing this problem by discovering new technologies and other approaches that could greatly accelerate the process of developing and deploying solutions that can be used by all translational researchers. To this end, one of the projects the center is focusing on is TRND Relaxin FIU project which has as goal to perform medicinal chemistry to measure type 1 collagen in a human cell line as a marker for inflammation. The HTRF Pro-collagen Type 1 (human) kit for the TRND Relaxin FIU project will continue research to support medicinal chemistry efforts. The goal of the project is to improve potency and PK properties of the current lead agonist of the relaxin receptor tough medicinal chemistry. In order to support medicinal chemistry efforts, we need to examine the compounds' effects on a cell based model for fibrosis by assaying type 1 collagen production. In order to have high enough toughput, the assay needs to be homogenous and compatible with miniaturized 1536-well format. This HTRF kit, or its equal, is required since much of the data has already been generated. Purpose and Objectives The purpose of this potential acquisition is to obtain Assay Reagent kits on a Brand-Name-or-Equal basis to the HTRF Pro-collagen Type 1 (human) kit as identified herein that is manufactured by Cisbio US, Inc., located at 135 South Road, Bedford, MA 01730-2204.
