The National Cancer Institute (NCI), Center for Cancer Research (CCR), Laboratory of Receptor Biology and Gene Expression (LRBGE), Optical Microscopy Core (OMC) plans to procure a Brand Name or Equal Intelligent Imaging Innovation Lattice LightSheet System: 4 color with two cameras and an analysis workstation.
This is a combined synopsis/solicitation for commercial items, prepared in accordance with format in FAR 12.6 as supplemented with additional information included in this notice. This announcement constitutes the only solicitation and a separate written solicitation will not be issued. This solicitation: No. 75N91019Q00057 includes all applicable provisions and clauses in effect through FAR FAC 2019-03 (June 2019) simplified procedures for commercial items.
The North American Industry Classification System code is 334516 and the business size standard is 500 employees.
Only one award will be made as a result of this solicitation. This will be awarded as a Severable Firm Fixed Price type contract.
BACKGROUND
The National Cancer Institute (NCI), Center for Cancer Research (CCR), Laboratory of Receptor Biology and Gene Expression (LRBGE) Optical Microscopy Core (OMC) requires the purchase of a Brand Name or Equal Lattice Light Sheet (LLS) Imaging System. This is a unique state-of the art system providing unprecedented spatiotemporal resolution for a long-term 3D non-photodamage non-photobleaching imaging. This translates into a quantitative and qualitative increase in information. Lattice Light Sheet state of the art technology surpasses the traditional techniques of live imaging such as laser scanning and spinning disc confocal technology as well as the other technologies of light sheet imaging. The LLS technique is widely applicable to the wide range of specimens from molecules to tissues and it will benefit numerous studies of cancer malignancy at CCR and NCI. Purchase of this system will be indispensable to the mission of NIH.
PRODUCT FEATURES/SALIENT CHARACTERISTICS
The following product features and salient characteristics apply to this acquisition:
· Optical Lattice Light Sheet Illumination Technique Based on Annular Mask Generating Parallel Arrays of narrow bessel light beams with adjustable light sheet thickness that can be scanned through the sample at high speeds reducing photodamage and photobleaching;
· Light Sheet Thickness, adjustable light sheet thickness and length with a minimum thickness of 0.4μm at 50μm length;
· Dither-mode LLS Resolution with improved Z: 230 x 230 x 370nm @ 1.1 NA;
· Super Resolution Lattice Structured Illumination (SIM): 150 x 230 x 280nm @ 1.1 NA;
· Detection Optics: water dipping objective, 2.0mm WD, 62.5x total magnification, which can be used without any material between the sample and objective;
· Illumination Optics: custom-made water dipping objective, 3.7mm WD, which can be used without any material between the sample and objective;
· Transmitted Light Imaging: LED light source and dedicated camera for location of cells;
· High Power Lasers, (> 300 mW): 405 350mW, 488 500mW, 560 500mW, 642 500mW;
· Two (2) Camera for Two-Channel Simultaneous Imaging: Hamamatsu ORCA-FUSION sCMOS camera with 80% QE and quiet scan 0.7e read noise, cooled to -5C, 2304x2304 pixels. The larger size of the detector on the ORCA-FUSION also results in a larger field-of-view. The reduced read noise compared to other cameras, such as the ORCA-FLASH 4.0 V3, is crucial for imaging samples with low numbers of photons. Alternative camera would be Photometrics Prime BSI, which has a read noise of 1.0 e.
· Chamber: Temperature and CO2 control with perfusion capabilities
· Specimen Mounting: Standard, non-proprietary chambers or slides, such as horizontally-oriented 5mm round glass coverslips which could also be imaged on other microscopes.
· Computer with Dual 10-Core, 2.4GHz processors, 128GB RAM, 8GB graphics card, 1TB OS SSD and 6TB SSD array, 10GbE (copper) Adapter
· Additional Processing Workstation with the Analysis Computer; Dual 10-Core, 2.4GHz processors, 128GB RAM, 8GB graphics card, 1TB OS SSD and 6TB SSD array, 10GbE (copper) Adapter
· Storage Solutions with DDN® unified storage systems for direct full-speed acquisition and analysis for the terabyte-size images, with a minimal storage size of 300TB. DDN systems utilize a BioScaler GPFS file system and are easily expandable to multiple petabytes.
· Lattice Tune-Up Capacities: Cameras in image and Fourier space to inspect the lattice and annular mask for tune up.
- Image Processing and Display: GPU accelerated deconvolution that can work from either a measured or theoretical PSF. 3D visualization software that can import multi-terabyte datasets and display them in a few seconds.
· Open design allowing modifications for the future incorporation of adaptive optics.
· All shipping, delivery and installation costs must be included in the quote. NCI will not be responsible after award is made, if shipping costs are not included.Contracting Office Address
