Item #                     Description                                             Qty.
001.    Task 1: Generate a single transgenic Pb line for                1 EA.
           Pf E140 antigen.
          a) A single transgenic Pb line expressing PF E140.
          b) Genotype data confirming the repplacement of E140 gene
          c) Data confirming the phenotype of the chimeric
sporozoites indicating its viavility and infectivity in DC1 mice.

002.  Task 2: Generate a double transgenci Pb line for Pf E140     1 EA.
         and  Pf Falstatin antigens.
         a)  A double transgenic Pb line expressing Pf E140 and
              and Pf Falstatin antigens.
         b)  Genotype data confirming the replacement of
              e140 and Falstatin genes.
         c)  Data confirming the phenotype of the chimeric
sporozoites indicating theirs viability and infectivity in CD1
              mice.
003.  Task 3: Generate a double transgenic Pb line                        1 EA
for Pf e140 and Pf CSP antigens.
         a) A double transgenic Pb line expressing Pf E140 and
             CSP antigens.
         b)  Genotype data confirming the replacement of E140 and
              CSP genes.
         c)  Data confirming the phenotyp of the chimeric sporozoites
indicating their viability and infectivity in CD1 mice.
004.  Task 4: Generate a double transgenic Pb line                          1 EA.
for Pf Falstatin and Pf CSP antigens.
         a)  A double transgenic Pb line expressing Pf Falstatin
              and Pf CSP antigens.
         b)  Genotype data confirming the replacement of Falstatin
              and CSP genes.
         c)  Data confirming the phenotype of the Tr Parasite
indicating its viability and infectivity in CD1 mice.
005.  Task 5: Generate a triple transgenic Pb line for Pf E140,            1 EA.
Pf Falstatin, and Pf CSP antigens.
         a)  A Pb line that express all three antigens: Pf E140,
Pf Falstatin and Pf CSP antigens by replacing Pb Falstatin
and Pf E140 genes by their Pf ortholog genes on a Pf
CSP transgenic Pb parasite. The CSP transgenic line
has been developed at LUMC by Dr. Shahid Khan.
         b)  Characterize the genotyp and phenotype of the triple
Tr line to confirm the correct replacement and the viability
of the parasite to infect CD1 mice.
