ADMINISTRATIVE INFORMATION

MULTI-OMICS CHARACTERIZATION OF COLD REGIONS SAMPLES

BLANKET PURCHASE AGREEMENT (BPA)

1. DESCRIPTION OF AGREEMENT.

The Biogeochemical Sciences Branch at the Cold Regions Research and Engineering Laboratory (CRREL) conducts various biological and chemical experiments on plants, microbes, and environmental samples. These experiments often require advanced multi-omics characterization of samples to identify biomarkers associated with biogeochemical processes.

The Contractor shall perform genomic, transcriptomic, proteomic, and/or metabolomic analysis of biological samples to reveal genetic pathways and organism functions. This agreement seeks to establish a Master BPA to provide up to 5 years of these services including analysis, data transfer, and bioinformatics as requested.

A BPA is a simplified method of filling anticipated repetitive needs for supplies or services by establishing "charge accounts" with qualified sources of supply. Pursuant to Federal Acquisition Regulation (FAR) 13.303, Blanket Purchase Agreements (BPAs), the Contractor agrees to the identified terms and conditions for a BPA to provide the required multi-omics sample services exclusively with the ERDC.

The primary aspect of this requirement shall include multi-omics analysis of plant, microbial, or environmental samples. This includes metabolomics, proteomics, transcriptomics, and genomics. Additional genomics services may also be requested for DNA library construction or gene synthesis.

Receive samples and perform quality assessment Generate omics data via metabolomics, proteomics, transcriptomics, and/or genomics services Process data and perform quality control Deliver raw data and description of instrument parameters used to collect data Perform data analysis

If and when requested, the vendor shall provide services in accordance with the specific requirements for the individual BPA Call/Order which may include, but are not limited to, the following broadly encompassing categories. It is not necessary for each entity to be capable of performing every service listed in this BPA list;

individual BPA calls will vary and may include one, some, or all of these services.

2. EXTENT OF OBLIGATION.

The Master Blanket Purchase Agreement (BPA) does not obligate any funds. The Government shall be obligated only to the extent of authorized purchases made via a properly executed BPA Call/Order.

3. PURCHASE LIMITATION.

The Government estimates, but does not guarantee, that the total volume of purchases through this agreement will be $500,000 over a 5-year period. Individual purchases shall not exceed that amount. There are no guaranteed minimum or maximum order limits established and no guaranteed dollar amount of expenditures with the vendor.

The Master BPAs will expire in five (5) years from date of award or total purchase reaching the BPA limit, whichever occurs first.

Delivery of results shall be FOB Destination delivered electronically to CRREL.

Actual purchases will be made via properly executed BPA Call. These Calls/Orders shall be initiated at the time the services or supplies are needed by an authorized representative of the Contracting Office.

4. TERMS AND CONDITIONS.

a) The Government shall be obligated only to the extent of authorized calls placed against this Blanket Purchase Agreement.

b) The prices for individual calls shall be obtained from the respective BPA holder.

Interested vendors need only to provide a response (letter format) which verifies the company’s interest and capabilities in providing the services outlined above, and enough information to provide CRREL with confirmation that the company can provide the requested services.

Interested vendors shall provide capability statements that include their company’s website address, price list, or catalog, if available, demonstrating representative pricing for supplies and/or services offered. The Government recognizes that no single vendor may be able to provide all requested services; therefore, multiple vendors may be selected as BPA holders.

There is no requirement for a “pricing bid” associated with this action.

Interested vendors response shall include the company Cage Code and/or Unique Entity Number (UEI).

The Master BPA will expire five (5) years from date of award or total purchases reaching the BPA limit, whichever is earlier.

Responses shall be submitted electronically to: Tomeka.J.McNeely@usace.army.mil

In order to be eligible, vendors must be actively registered in the System for Award Management (SAM), have no active exclusions, and complete the FAR & DFARS representations and certifications. For more information on the SAM, review the SAM website at https://www.sam.gov.

PERFORMANCE WORK STATEMENT

Multi-omics Characterization of Cold Regions Samples

1. INTRODUCTION: The Biogeochemical Sciences Branch at the Cold Regions Research and Engineering Laboratory (CRREL) conducts various biological and chemical experiments on plants, microbes, and environmental samples. These experiments often require advanced multi-omics characterization of samples to identify biomarkers associated with biogeochemical processes.

2. OBJECTIVE: The objective of this Blanket Purchase Agreement (BPA) requirement is to obtain Multi-omics Characterization of Cold Regions Samples from vendors able to support CRREL requirements. The Contractor shall perform genomic, transcriptomic, proteomic, and/or metabolomic analysis of biological samples to reveal genetic pathways and organism functions. Other ERDC labs may also use this BPA with coordination of the CRREL COR.

3. MAJOR REQUIREMENTS: In order to accomplish the work under this requirement, it shall be necessary, based on the individual practices and policies of the requested centers, for the contractor to provide the following tasks.

Specific BPA calls may request only certain portions of these requirements or may request all of these requirements.

For example, a BPA call may request one, some, or all of the analysis categories under Task B. It is acceptable that vendors participating under this Master BPA may only be capable of providing some of the services under Task B.

Also, individual BPA calls may or may not ask for Data Integration and Analysis (Task E).

Task A. Sample Reception and Quality Assessment:

Receive and document all biological samples provided by CRREL Perform initial quality assessment of samples (e.g., volume, mass). Report any issues immediately.

Task B. Omics Data Generation:

CRREL will identify which omics analysis is desired at the time of the proposal call. One call may not include all sample analyses, the call will specify which one we want.

Genomics: Perform Sanger sequencing, amplicon sequencing (250 bp paired-end reads), shotgun metagenomics sequencing, or whole genome sequencing (WGS) at desired sequencing depth.

Must use Illumina sequencing platform for amplicon and shotgun metagenomics or Nanopore sequencing platform for WGS.

Transcriptomics: Perform RNA sequencing (RNA-Seq) with 250 bp paired-end reads and a target of 30 million reads per sample.

Proteomics: Perform targeted and untargeted proteomics via Liquid Chromatography-Mass Spectrometry (LC-MS/MS)

Metabolomics: Perform targeted and untargeted metabolomics via Gas Chromatography-Mass Spectrometry (GC-MS) or Liquid Chromatography-Mass Spectrometry (LC-MS) for comprehensive metabolome profiling.

Conduct DNA Library construction as requested Conduct gene synthesis as requested

Task C. Data Processing and Quality Control:

Perform raw data processing for each omics platform (e.g., demultiplexing, read alignment, peak detection).

Implement rigorous quality control (QC) procedures to identify and remove low-quality data.

Normalize data to account for technical variations.

Task D. Deliver Raw Data:

Deliver raw data as appropriate for each omics analysis

Provide instrument parameters for each omics analysis run

Task E. Data Analysis:

Bioinformatics services may be requested by the proposal call Integrate the processed multi-omics datasets using appropriate bioinformatics tools and statistical methods.

Perform differential expression/abundance analysis to identify significant changes between treatment groups.

Perform pathway enrichment analysis to identify biological pathways significantly altered.

Conduct correlation analysis to identify relationships between different omics layers.

Perform statistical analysis to ensure the robustness and significance of findings (e.g., p-value adjustment, false discovery rate control).
