Scope of Work
Title: CRISPR- (lox)
The contractor offers needed services that produce tissue-specific, homozygous knockout mice. The technology has to be proven to be advantageous over conventional gene targeting or mouse embryonic stem (ES) cell based approaches. They are an organization that is focusing on mammalian genetics research to advance human health research. Thru their services, contractor aids in discovering precise genomic solutions for disease, and empowers the global biomedical community in the shared quest to improve human health.
Included System Component (s):
The contractor provides the following required workflow: They will produce brain tissue specific gene knockout process. More specifically, they will first generate two (2) different LoxP alleles (i.e. conditional knockout strains) using the technology. They will then cross these conditional knockout strains to selective Cre strain (TBD), to achieve tissue specificity in brain regions (hippocampus or amygdala). Lastly, they will perform necessary subsequent breeding, such as expanding the colony to meet the delivery goals. They will meet a delivery goal of a one –time shipment of twenty (20) strains tissue-specific, homozygous knockout mice that are male and between 4-6 weeks of age.
Period of Performance: Shipment will be available approximately 26 months from the initiation date.
Deliverables:
1. Generation LoxP alleles (i.e. conditional knockout strains) using CRISPR technology
2. Cryopreservation of N0 founders
3. Cryorecovery of Cre strain
4. Breeding
