Performance Work Statement (PWS) U.S. Army Center for Environmental Health Research
Global Metabolomics and Quantitative Fatty Acid Composition of Cardiac Tissue and Plasma from Exertional Heat-Stroked Mice
Vision Statement Conduct high quality metabolomic and fatty acid content analysis on cardiac tissue and plasma specimens
1 Introduction The U.S. Army Center for Environmental Health Research (USACEHR) requires metabolomic services to support our heat stroke research. These services (1) provide us the ability to analyze and catalogue the biochemical, fatty acid, and metabolic changes in cardiac tissue and plasma from mice exposed to exertional heat stress, (2) provide the basis to determine whether future specialized analysis and techniques will be required, and (3) generate data to integrate with physiological and various "-omics" data related to the project.
1.1 Mission
The mission of USACEHR is to provide diagnostic and prognostic tools for environmental and occupational health surveillance. As part of a new effort to increase the use of a systems biology approach across the U.S. Army Medical Research and Materiel Command (USAMRMC), USACEHR is expanding its capabilities in genomics, proteomics and high performance computing. Systems biology offers tremendous opportunities to accelerate the pace of discovery for biomedical applications through the use of biological experimentation that is tightly linked to mathematical models and a better understanding of biological networks and molecular biology. With support from USAMRMC, USACEHR hopes to increase its efforts in this exciting area while contributing to Command-wide applications of systems biology approaches.
1.2 Background
Heat stroke is a serious medical problem for military personnel in training and deployment in hot climates. Heat illness is a continuum of disorders ranging in severity from temporary to fatal. Under heat stress, central blood volume is shunted to the periphery to facilitate evaporative cooling. Eventually, diminished total blood volume provokes hyperthermia and threatens the maintenance of blood pressure. When cardiovascular needs take priority over body temperature, severe hyperthermia and heat stroke syndrome develop. As a result, detrimental cellular and molecular changes occur that lead to a cascade of events including acidosis, ischemia, hypoxia, cellular fatigue, CNS dysfunction, and decreased blood pressure. Profound derangements in these physiological systems trigger endotoxemia, systemic inflammatory response syndrome (SIRS), disseminated intravascular coagulation (DIC), and ultimately multiorgan dysfunction syndrome (MODS). Meta-analysis of systems data and organ injury will be examined to determine return to duty (RTD) candidate biomarkers. Data generated will be integrated into a systems biology data cube relevant to military medical disorders and syndromes at the Advanced Biomedical Computing Center (ABCC), which will serve as an integrated platform enabling the discovery of prognostic biomarkers, driver pathways and development of predictive mathematical models.
2. Performance Objective:
The contractor shall provide non-personal services carrying out analytical studies to compare and report the statistically significant biochemical differences between control cardiac tissue or plasma and corresponding experimental cohorts. The contractor shall employ a fast and reproducible method directly reflecting biochemical events through the determination of changes in the levels of metabolites (including complex lipids and fatty acids) in biological samples like cardiac tissue and plasma, owing to physiological stimuli or genetic modification. After monitoring these biochemical changes following physiological stress, the contractor shall perform statistical analysis and report biochemical interpretation of results.
3. Scope of Work:
The contractor shall provide analysis of 70 mouse cardiac tissues (left ventricle) and the corresponding 70 plasma specimens from each animal. The project will consist of customized sample preparation, evaluation of samples for the presence of metabolites utilizing LC/MS and GC/MS mass spectrometry technologies, data processing, statistical analysis, and preparation of a final report. Both global, semi-quantitative metabolomics and a fully quantitative profile of concentration and fatty acid composition ofneutral and polar lipid classes shall be performed on both the cardiac tissue (left ventricle) and the plasma specimens.
| 1) | The contractor shall provide advice on preparing the cardiac tissue (left ventricle) samples while preserving the metabolic entities for analysis. |
| 2) | The contractor shall deliver a report of the relative concentration profile of the small-molecule entities extracted from the cardiac tissue and plasma and the absolute quantitative profile of the concentration of fatty acid composition of neurtal and polar lipid classes. The small-molecule entities shall be identified by liquid chromatography/mass spectrometry and gas chromatography/mass spectrometry detection systems (i.e., endogenous compounds, xenobiotics, and their metabolites, as detected samples and detected by LC/MS and GC/MS (mass spectrometry) systems.contractor shall deliver a report of the relative concentration profile of the small-molecule entities extracted from the cardiac tissue and identified by liquid chromatography/mass spectrometry and gas chromatography/mass spectrometry detection systems (i.e., endogenous compounds, xenobiotics, and their metabolites, as well as biomolecules of indeterminate structure). |
| 3) | The contractor shall deliver a statistical analysis of the resulting data, including ANOVA with contrasts and random forest analyses. |
| 4) | The contractor shall provide a biochemical interpretation of selected, statistically significant, named compounds (i.e., known chemical structure) using pathway analysis tools. The contractor shal integrate the biochemical interpretation into a final report summarizing the statistical and biological conclusions of the study. |
| 5) | The contractor shall provide access to a Client Portal tool which incldes heat maps, charts, pathway enrichment analysis, and an annotated Network View of the data. |
| 6) | The contractor shall provide access to Web-based tools for intuuitive data visualization of lipids identified in the quantitative profile of concentration and fatty acid composition of neurtal and polar lipid classes. |

7. Deliverables:
The contractor shall deliver the final report—including all statistical analysis and biochemical interpretation described in the above Scope of Work—within three (3) months after the receipt of the samples.
Quality Assurance Surveillance Plan (QASP) U.S. Army Center for Environmental Health Research
Global Metabolomics and Quantitative Fatty Acid Composition of Cardiac Tissue and Plasma from Exertional Heat-Stroked Mice
Purpose This Quality Assurance Surveillance Plan (QASP) provides a systematic method to evaluate contractor performance.
This QASP does not detail how the contractor accomplishes the work. Rather, the QASP is created with the premise that the contractor is responsible for management and quality control actions to meet the terms of the contract. It is the Government’s responsibility to be objective, fair, and consistent in evaluating performance. In addition, the QASP should recognize that unforeseen and uncontrollable situations may occur.
This QASP is a “living document” and the Government may review and revise it on a regular basis. However, the Government shall coordinate changes with the contractor. Updates shall ensure that the QASP remains a valid, useful, and enforceable document. Copies of the original QASP and revisions shall be provided to the contractor and Government officials implementing surveillance activities.
FAR 52.212-4(a) Inspection/Acceptance applies to this effort.
Methodologies to Monitor Performance:
In an effort to minimize the contract administration burden, the Government will use simplified methods of surveillance to evaluate contractor performance. The primary methods of surveillance will document the degree to which the contractor meets the agreed-upon tasks outlined in the PWS. The acceptable quality levels (AQL) are located in Table 1: Required performance Metrics for Contractor Performance. The Contracting Officer’s Representative (COR) will evaluate the services required by each task to ensure compliance.
Table 1: Required Performance Metrics for Contractor Performance
Required Task Performance Standard
| Method of Surveillance | Acceptable Quality Level | Incentive | ||
| Provide advice on preparing the cardiac tissue (left ventricle) and plasma samples while preserving the metabolic entities for analysis | Contractor will report that samples have been run successfully and X number of metabolites were seen | The COR will discuss if sample size is adequate to identify a broad range of metabolites | Approximately 200 metabolites should be detected | Favorable past performance rating |
| Deliver a report of the relative concentration profile of the small-molecules and an fully quantitative profile of the concentration of fatty acid composition and neutral and polar lipid classes that have been extracted from the samples and detected by mass spectrometry | Contractor will report if hormones have been identified along with metabolites | The COR will discuss if sample size is adequate to identify hormonal responses | Type of hormonal responses will be assessed and related to exposure used | Favorable past performance rating |
| Deliver a full statistical analysis of the resulting data | Contractor will match control/test groups/ pairs and report relative changes due to exposure and recovery | The COR will correlate report of relative changes to any published results, if any exist | A series of related changes should imply pathway regulation | Favorable past performance rating |
| Provide a biochemical interpretation of selected, statistically significant, named compounds using pathway analysis tools | Contractor will provide information as a final report | The COR will integrate the reported findings with other pan-omic results for consistency | The significant changes will provide potential druggable targets | Favorable past performance rating |
