1.0 TITLE: The contractor shall be required to perform In silico screening/molecular modeling and allow use of their Linux cluster for x-ray crystal structure data refinement for therapeutic and vaccine development against bacteria, parasites, and viruses.

2.0 BACKGROUND: The mission of the Walter Reed Army Institute of Research (WRAIR) is to conduct biomedical research that is responsive to Department of Defense and U.S. Army requirements and delivers life saving products including knowledge, technology, and medical materiel that sustain the combat effectiveness of the war fighter. The Division of Bacterial and Rickettsial diseases (BRD) implements the mission of the WRAIR by developing medical solutions for protecting U.S. service members against pathogenic bacteria. The Department of Wound Infection within BRD, targets pathogens commonly found in wound infections, and this Department also houses the WRAIR X-ray Crystallography Facility which researchers utilize for structure-based drug design in therapeutic and vaccine development against bacteria, parasites, and viruses.

3.0 SCOPE OF WORK:

The proposed work to be performed by the contractor involves virtual screening of commercially available database for antimicrobial and anti-malarial therapeutic discovery, docking of therapeutic candidates into protein targets and performing molecular modeling.

3.1 Detailed tasks:

3.1.1 Perform in silico screening of commercially available chemical databases using the crystal structures of bacterial therapeutic targets from the MEP pathway, a malaria (parasite) therapeutic target, wound infection bacterial therapeutic targets and Shigella therapeutic targets.

3.1.2 Perform molecular modeling to study the protein:lead compound interaction. The protein crystal structures for in silico studies will be provided by the PI/COR, (WRAIR).

3.1.3 The contractor must be able to integrate biological, chemical, and structural data for each of the therapeutic target compounds.

3.1.4 The contractor must have linux operating systems or Mac OS X for running x-ray structure refinement programs in PHENIX and able to write computer scripts such as PERL, linux batch script, commercial molecular simulations software.

3.1.5 The contractor should be within 10 miles of WRAIR because the COR has to travel to the offsite location to use their Linux cluster to refine the x-ray crystal structures of potential therapeutic targets on a frequent basis.

The contractor would provide approx 5 inhibitors per week to the COR.

The COR would then grow these crystals in complex with the inhibitors.

To study the successful crystals grown the COR needs to commute to the contractor location twice a week to process the data using their linux cluster.

3.2 Qualifications

3.2.1 The contractor must provide service that should possess a doctorate professional caliber with years of experience/knowledge in Biophysics or related field with computer modeling background to accomplish the tasks mentioned in the scope of work.

4. DELIVERABLES:

4.1 A quarterly report describing all work done in support of this PWS will be delivered by the 12th of January, April, July, and October to the PI/COR, (WRAIR).

4.2 Structures of compound leads, modifications to lead compounds, suggested mutations to proteins and vaccine candidates will be delivered to the COR via email within 5 days of identification.

5. QUARTERLY DEPARTMENTAL PROJECT MEETINGS:

The contractor will be required to attend meetings on site at WRAIR at a minimum 4 times a year.

6. GOVERNMENT FURNISHED ITEMS:

6.1 An official use computer system (desktop) will be made available for off-site support

6.2 The Government shall provide the software such as MOE, ChemNavigator, Amber, Pymol to accomplish the task mentioned in the scope of work.

6.3 Government will replace Government-provided computer if the computer is not functional.

7. WORK SCHEDULE AND LOCATION: The contractor shall work at their location within a 10 mile radius of WRAIR, 503 Robert G Avenue, Silver Spring, MD.

8. PLACE OF PERFORMANCE: Contractor’s site.

9. PERIOD OF PERFORMANCE: 1 year from the time the contract is awarded .
