SALIENT CHARACTERISTICS
The laser microdissection microscope system is required for the isolation of cells from histological sections from healthy vs diseased tissue biopsies including archived samples. This is necessary for conducting translational research studies aimed at understanding diseases in patient derived samples.
A. Must capture/collect specimen by gravity for increased collection speed and avoidance of sample contact contamination.
B. The system must operate with the shortest user training and effort. Novice users should be capable of collecting high fidelity samples for experimental use within less than 1 day training.
C. Samples can be cut and collected into inexpensive and commercially available plastics. Collection device are flexible to accommodate sample placement into 0.5mL or 0.2mL caps, 8-well strips from a 96-well plate, dishes for cell culture, chamber slides.
D. Fully automated upright Compound microscope for greater laser cutting flexibility, speed and resolution with microscope stand with fully motorized controls and software interface.
E. Intuitive setup of experiment conditions for acquisition of up to 8 multi-channel sequences for seeming less multi-dimensional images with multi-channel, time lapse, and z-stack.
F. Capable of storing personal settings and imaging settings from a previously acquired image.
G. Requires adaptive focus control for best in the field for maintenance of focus. Software that has move and cut feature for real time control of laser cut by mouse, allowing for simple cutting and reproducibility.
H. System that works independently as full imaging system with image acquisition and analysis software that can be used to also quantify histochemical staining’s.
I. Software components and features must include autofocus, serial section cutting automated collection and positioning of collection tubes, automated inspection more, multi-cutting over the entire slide, and export of shape list data to MS-Excel.
J. Color camera with USB3 connection, 2/3” interline progressive scan CCD sensor and minimum of 2.8 megapixel resolution.
K. Includes all connection adapters and mounts for porting all peripherals.
L. 355nm pulsed diode laser capable with adjustable cutting speeds, laser power, laser aperture, and laser offset. Must be adjustable for cutting thick, thin, soft, and hard specimens.
M. Sample flexibility cutting range from 5-200+ microns thick, live cell dissection and live specimen dissection.
N. Triple slide stage holder and four tube collectors for high throughput specimen collection.
O. No limitation of surface area for amount of dissectate collected. The system uses the entire volume of the specimen to maximize the collection of material from rare and precious samples.
P. Full HD LCD monitor at least 24” with microscope manufacturer compatible PC workstation that uses 64bit Windows 10 or later with a minimum of 64GB RAM, 512GB SSD hardrive, 4TB storage hard drive, 4GB dedicated graphics card, and DVD writer.
Q. Alignment-free external lightsource with attenuator and fast shutter.
R. Dedicated lenses for high UV throughput to include 5x, 10x, 20, and 40x cutting objectives and 63x and 100x oil objectives for in depth analysis.
S. Quick simultaneous live cutting within fluorescence that maintains the ability to watch the dissection in real time during fluorescence imaging.
T. Built in internal filter wheel for fluorescence excitation with one triple fluorescence cube for three independent channels. Must include intensity and excitation manager for automated software control. Blue, green and red fluorescent channels included.
U. Ability to adjust power, aperture, pulse frequency, speed, UV-offset, and specimen balance settings for each objective, for each sample.
V. High stability laser that requires no warm up time or cool down between experiments or users.
W. Anti-vibration table (air table) specified by system manufacturer.
X. Able to provide direct manufacturer representation and support in the Seattle/Tacoma metropolitan areas to provide initial set up, training and technical support.
