STATEMENT OF REQUIREMENTS

TITLE: Biocapacitance Probe System

I. BACKGROUND

The Bioprocess Measurements Group and the Biomolecular Structure and Function Group within the Biomolecular Measurement Division of the National Institute of Standards and Technology are responsible for the improving measurement science and generating reference data with a focus on biopharmaceutical manufacturing. For bioreactors, there is a need for increasing online data measurements for quality control. There are very few measurements that can assess cell viability in the reactor. Biocapacitance measurements can provide this information. Traditionally, single measurement frequency is used, but there is growing interest in the capability for multifrequency measurements to provide more detailed information on the state of cells in the bioreactor.

II. SCOPE OF WORK

The Bioprocess Measurements Group and the Biomolecular Structure and Function Group require a biocapacitance probe system for research into advancing measurement capabilities for bioreactors. The goal is to use multi-frequency analysis to provide online data about cell viability, and state of cells in the bioreactor. Because we are conducting research on the fundamental measurement principles, we require a biocapacitance system with maximum frequency range and full access to measured parameters.

III. SPECIFICATIONS

The Contractor shall provide one (1) biocapacitance probe system that meets the following minimum technical requirements.

The item must be new and commercially available. Used or remanufactured equipment will not be considered for award.

Technical Requirements:

•The system shall have the ability to detect less than 300,000 cells/ml at the cell size of 12 mm
•The system shall have a probe port diameter 12 mm, probe length 100-250 mm.
•The system shall include appropriate fittings to allow probe height in bioreactor to be adjusted over a range of at least 100 mm.
•The system shall have a measuring frequency range of at least 50 kHz to 20 MHz
•The system shall have at least 25 frequency values available
•Frequency scanning algorithms must be open-source
•For non-frequency scanned measurements, the frequencies used can be set according to sample type (cell culture, bacteria, microbes)
•The probe must be fully autoclaveable
•The system shall include all electronics, power supply, and software to drive probe.

Installation is not required.

IV. DELIVERABLES

Deliverable: The contractor shall deliver to the NIST in Gaithersburg, MD one biocapacitance probe system meeting all the requirements listed above.

Standards for Acceptance of Deliverables: The NIST Technical Point of Contact will inspect the biocapacitance probe system upon delivery to confirm it operates within specifications. The warranty shall not commence until final acceptance has been granted by the Government. Should any deficiencies be noted prior to final acceptance, the Contractor will be notified and provided the opportunity to correct all deficiencies, at their expense, in accordance with the commercial terms and conditions of the contract.

Delivery: Delivery terms will be FOB Destination. FOB Destination means: The contractor will pack and mark the shipment in conformance with carrier requirements, deliver the shipment in good order and condition to the point of delivery specified in the purchase order, be responsible for any loss of and/or damage to the goods occurring before receipt and acceptance of the shipment by the consignee at the delivery point specified in the purchase order; and pay all charges to the specified point of delivery. The contractor shall deliver the biocapacitance probe system to:

Richard Cavicchi Biomolecular Measurement Division National Institute of Standards and Technology Room B312, Building 221 100 Bureau Dr.

Gaithersburg, MD 20899-8362

V. Delivery Date The delivery shall be 60 days from the date of award.

VI. WARRANTY

The Contractor shall provide, at a minimum, a one-year warranty for the equipment. The warranty shall cover parts and labor. The warranty shall commence upon successful completion of delivery.
