STATEMENT OF WORK

Sugarcane Chromosome Karyotype Analysis

Introduction: As part of the sugarcane breeding activities at the Sugarcane field station in Canal Point, parental fertility is very important to produce high quality viable seeds. Since sugarcane is highly polyploid with aneuploidy, it would be beneficial to have the parent’s cytological information available.

Scope: The Contractor shall perform the cytological analysis on sugarcane parents to determine their ploidy levels and identify molecular markers.

Technical Requirements:

· high-resolution metaphase chromosome preparation technology

· fluorescence in situ hybridization

Period of Performance: Within 90 days after award.

Government Furnished: Access to the facility during normal business operations (Monday through Friday, 7:30 am to 4 pm)

Performance Requirement Measures:

· Count chromosome numbers using high-resolutions chromosome preparation technique.

· Develop Fluorescence in situ hybridization for parents.

· Provide data and analytics.

Service Description:

Chromosomes karyotype analysis and FISH

• Material: Sugarcane knots to be provided by customer (10 sugarcane knots will be great, if not, provide as many as possible)

• Root tips will be collected from the germinated sugarcane knots• Probe: Genome DNA from one genotype

- Genome DNA will be extracted from the roots

• Deliverable

- Detailed report of methods and results;

- Complete raw data set;
