About this file

This is a Statement of Work (SOW) for a sole-source contract to provide an assurance plan for maintenance and repairs of a 10x Genomics Chromium X single cell capture system (Serial Number CXVG11667) located at the NIH Porter Neuroscience Research building in Bethesda, MD. The contract requires the contractor to provide preventive maintenance, replacement of reagents lost during failed runs, and instrument replacement due to unrecoverable errors.

The contractor must provide factory-trained field service representatives to perform maintenance between 7am-6pm Monday through Friday, including examination of functioning parts, repairs, calibration, and complete dismantling when necessary. The period of performance is for a base period plus four option years. The system is used by the Molecular Neurobiology Section at NINDS for sequencing regions of open chromatin and gene expression in individual cells for neural regeneration research. Key requirements include maintaining the equipment in operable condition, minimizing downtime, and ensuring compatibility with existing laboratory protocols. Service personnel must be certified by 10x Genomics as the Original Equipment Manufacturer.

Version 11-30-2016-1 FAR Part 11—Describing Agency Needs

STATEMENT OF WORK (SERVICES)

GENERAL INFORMATION

Title of Project:

Assurance Plan for 10x Genomics Chromium iX/X (Serial Number CXVG11667)

Statement of Need and Purpose:

The purpose of this acquisition is to acquire an assurance plan for maintenance and repairs of the 10x Genomics Chromium X single cell capture system. The Chromium X is used routinely by the members of the Molecular Neurobiology Section in NINDS. This contract is required in order to maintain this research equipment in operable condition, provide replacement of reagents lost during a failed run, and ensure instrument replacement due to an unrecoverable error. The requirement is for three months and issued on a sole-source basis from 10x Genomics.

Background Information and Objective:

The National Institutes of Health (NIH) is the nation’s leading medical research agency and the primary Federal agency whose mission is to seek fundamental knowledge about the nature and behavior of living systems and the application of that knowledge to enhance health, lengthen life, and reduce illness and disability, conducting, supporting and making medical discoveries that improve people’s health and save lives.

The mission of the Molecular Neurobiology Section is to identify the signaling pathways important for neural regeneration after injury. The lab currently uses mouse olfactory epithelium, which has robust regenerative capacity, as a model for neural regeneration. A primary approach to understanding mechanisms of neural regeneration is to sequence the regions of open chromatin, which correlate gene regulatory elements, in individual nuclei and the genes that are expressed in individual cells. The Chromium X device partitions cells or nuclei into individual “gel emersions” that allow for the generation and sequencing of their gene expression profiles (transcriptomes) and accessible chromatin profiles (epigenomes). The Chromium X has the capability of partitioning thousands of individual cells or nuclei from a single sample, which are then used to construct a single cell sequencing library.

The Chromium X single cell capture system is required to partition cells/nuclei for single cell sequencing libraries that are compatible with libraries generated in the Molecular Neurobiology Section over the past

3.5 years. The Chromium X requires routine preventative maintenance and an assurance plan that covers lost reagents due to a failed run. This contract will ensure minimal downtime due to hardware and/or software malfunction. Additionally, the contract is required to maintain this research equipment in operable condition for single cell/single nuclei partitioning (“captures’), provide for the replacement of lost replacement during failed captures, and prevent costly repairs.

Period of Performance:

The period of performance is for base plus four option years.

SCOPE OF WORK

General Requirements:

The contractor shall provide a qualified technician to provide maintenance as needed on the laboratory’s Chromium X system (Serial Number CXVG11667).

Independently and not as an agent of the Government, the Contractor shall furnish all the necessary services, qualified personnel, material, equipment, and facilities, not otherwise provided by the Government as needed to perform the Statement of Work below:

Specific Requirements:

Contractor shall provide all labor and replacement parts to maintain normal operation of the existing Chromium X. Service shall be provided between the hours of 7 am and 6 pm, Bethesda, MD local prevailing time, Monday through Friday, unless indicated otherwise as follows:
