Version 11-30-2016-1 FAR Part 11—Describing Agency Needs

STATEMENT OF WORK

GENERAL INFORMATION

Title of Project:

DNA extraction, genotyping and analytical services

Statement of Need and Purpose:

DNA extraction, genotyping services of DNA samples and analysis in support of research in Guillain-Barré syndrome (GBS).

Background Information and Objective:

Guillain-Barré syndrome (GBS) is a rare disorder where the muscles are attacked by the immune system causing peripheral nerve cell damage. The underlying cause is not fully understood. It has been observed that some individuals can develop GBS after having flu or other infections, or in days or weeks after receiving vaccination. Here, we aim to delineate the genetic risks that predisposes individuals to acquiring GBS. This knowledge will help in the stratification of individuals in patient care and vaccine recommendation and administration.

Period of Performance:

8 months

SCOPE OF WORK

General Requirements:

Independently and not as an agent of the Government, the Contractor shall provide all biological samples from GBS and healthy individuals and furnish all the necessary services, qualified personnel, material, equipment, and facilities, not otherwise provided by the Government as needed to perform the Statement of Work below.

Specific Requirements:

The Vendor shall perform automated DNA extraction on 1080 proprietary samples owned by Erasmus MC of Rotterdam. Vendor will provide QC results of an initial DNA extraction to the Government to confirm DNA quantity and quality for each sample.

Acceptable quantity of DNA is defined as a yield of 50ng per sample. Acceptable quality of DNA is defined as absorbance 260/280 ratio value equal to 1.8nm, and an absorbance 260/230 ratio value greater than 2.0nm.

Version 11-30-2016-1 FAR Part 11—Describing Agency Needs

After consultation with the Government, samples passing this criteria will then be approved for genotyping processing and genetic association analysis. The Vendor shall have access to and shall perform the array-based genotyping on extracted DNA using the Illumina Infinium iSelect 24x1 HTS Custom Beadchip. This custom beadchip is the Infinium Global Screening Array-24 v3.0 BeadChip (GSA MD version 3), as described at http://glimdna.org/global-screening-array.html. The Vendor shall provide processed quality control metrics and summary statistics of the analysis to the Government via upload to a secure system such as Google Cloud, SFTP, DNANexus, or similar within 26 weeks of receiving purchase order.

LEVEL OF EFFORT:

Vendor staff shall provide the assay quality control and results to the Government via secure file transfer.

DELIVERY OR DELIVERABLES

Minor allele frequencies and association analysis results for the 1080 samples.

OTHER CONSIDERATIONS

Travel:

The principal place of performance is the vendor’s work site. Travel for contractor employees will not be necessary.

Key Personnel:

Vendor shall provide a dedicated project manager to the government as a point of contact for the services.

Section 508—Electronic and Information Technology Standards:

The contractor shall comply with Section 508 of the Rehabilitation Act (29 U.S.C. § 794d), as amended by the Workforce Investment Act of 1998 (P.L. 105-220), August 7, 1998.
