About this file

This document is a Statement of Work (SOW) for a federal contract opportunity related to analyzing genetic data to identify pathways linking hypertension to dementia. The purpose is to replicate a previous analysis finding blood pressure pathways associated with cerebral small vessel disease.

The contractor will be required to have expertise in analyzing large genetic datasets, using statistical packages like R, PLINK, Python, and Lynx, and access to a suitable computing infrastructure. The analysis should be completed within 6 months, and the deliverable is a draft manuscript ready for journal submission. The government will review and approve the final analysis. This SOW is associated with a federal contract opportunity solicitation number 75N95024Q00329, issued by the Department of Health and Human Services' National Institute on Drug Abuse on behalf of the National Institute on Aging. The contract will be awarded on a sole-source, fixed-price basis to Queen Mary University London as the only source able to access the required dataset and analysis pipeline.

Version 11-30-2016-1 FAR Part 11—Describing Agency Needs

STATEMENT OF WORK (SERVICES)

(SOW)

GENERAL INFORMATION

Title of Project:

Key drivers of hypertension as a risk factor for dementia.

Statement of Need and Purpose:

The purpose of this requirement is to replicate a finding of genetic and related physiological pathways that drive the association of hypertension to brain pathology.

Background Information and Objective:

Dementia is a devastating and global public health problem with an urgency to identify public health interventions and therapeutics. It has been shown, in many different cohorts, that hypertension is a major, modifiable risk factor for dementia. A recent large scale multi-ethnic trial provided evidence that lowering systolic blood pressure leads to a reduction in a composite dementia/mild cognitive decline and in white matter lesions (a marker of small vessel disease), but no effect on total brain volume. These analyses were limited and give little insight into the underlying changes in the brain that may account for the cognitive benefit.

We have recently identified blood pressure pathways that are also significantly associated with brain- MRI traits. The strongest signals are from 8 pathways, which may be potentially important in explaining the underlying mechanisms through which hypertension effects small vessel disease [manuscript in progress: O. Meirelles, C. Cabrera, Z. Li, AK Gupta, M. Brown, MA. Nalls, MB Barnes, PB. Munroe, LJ Launer, HR Warren]. It is standard good practice have discovery findings based on genetics to be replicated in other studies.

The aim of this contract is to conduct data analysis and generate a report that includes data on the replication of this already existing analysis.

Period of Performance:

6 months

SCOPE OF WORK

General Requirements:

1.The Contractor shall furnish all the necessary services, qualified personnel, material, equipment, and facilities, not otherwise provided by the Government, as needed to perform the specific requirements below/

2. The contractor will data analyze different datasets and generate a report showing replication of an already existing pipeline and analysis showing blood pressure pathways are associated with cerebral small vessels disease [O. Meirelles, C. Cabrera, Z. Li, AK Gupta, M. Brown, MA. Nalls, MB Barnes, PB.

Munroe, LJ Launer, HR Warren (in progress)].

Specific Requirements:

To implement the analysis the contractor should have expertise and documented experience in conducting analyses with very large cohorts that have genome-wide data on all participants. To manage

Version 11-30-2016-1 FAR Part 11—Describing Agency Needs and analyze the genetic data, the contractor should provide evidence of expertise in the following statistical packages: R, PLINK, Python, and Lynx. In addition, the contractor must have immediate access to a computer infrastructure with an appropriate set-up to store and process large (GBs) datasets. The computer storage system should be large enough to store and analyze large (GB) genetic databases and output results; can process genetic datasets within a reasonable time; and meets nationally acceptable security and privacy standards. Data will be analyzed with a fixed set of variables used in the discovery analysis but may have to be reformatted/coded in the replication dataset to be compatible with the analysis pipeline. The contractor must have access to the pipeline that was used in the first exam and will be the basis for the replication analysis

A report of the data analysis results in an excel file will be provided to the NIA project officer.

GOVERNMENT RESPONSIBILITIES

The Government will review and approve the final analysis of data based on an approved plan.
