Statement of Work for Bacterial Artificial Chromosome (BAC) library creation using Advanced Random-sheared Technology (ART)
One of our research objectives is to identify soybean genes that confer resistance to a disease known as soybean rust. The soybean line ‘PI 605823’ is known to harbor a resistance gene called Rpp7. To accurately sequence the region of the soybean genome where Rpp7 is located, long (< 100 kb) contiguous fragments of the genome are required. This is typically accomplished through the creation of bacterial artificial chromosome (BAC) “libraries” where each BAC contains a portion of the soybean genome. BACs that contain the gene or region of interest are then identified and the DNA is sequenced. We have used this technology in the past to obtain information for other soybean varieties that have resistance to this disease. These BAC libraries have resulted in numerous data sets and multiple scientific publications.
The most efficient way to obtain a BAC library is to work with a company that specializes in this service. Constructing a BAC library is very time-consuming, expensive and requires more labor than we have available. We would like to obtain a BAC library constructed with ‘advanced random-shear technology’, which is a newer technology that provides better coverage of the genome than the traditional restriction enzyme method. This technology provides for a more useful product for our scientific studies.
We will ship the required amount of soybean leaves from the soybean line PI 605823 (Rpp7) on dry ice to the vendor and they will use that material to construct a BAC library. The company will use the advanced random shearing technology in the construction of the library. The company will provide a library that has an average insert size of approximately 120kb and 10X coverage of the genome. The company will provide the BAC library as individual clones in 384-well plates. The company will also prepare and provide plate-pools (where each well represents a single plate of BACs). The company will also identify at least one positive clone using oligonucleotide primer sequences that we will provide. The location of this positive clone will be provided in the final project report. The company will ship this library on dry ice to the given address and will also provide a complete project report and certificate of analysis.
The vendor will provide a step-by-step timeline on creating, screening, and shipping the BAC library. We propose a delivery timeline of 16 weeks, starting from the date the materials are received by the vendor.
