STATEMENT OF REQUIREMENTS
Liquid Chromatography (LC) Instrument with Fluorescence Detector
I. BACKGROUND
The National Institute of Standards and Technology (NIST), Material Measurement Laboratory
(MML), Biomolecular Measurement Division (BMD) is responsible for the development of chemical databases to accomplish NIST’s mission to carry out collection, compilation, critical evaluation, publication and sale of standard reference data in accordance with Public Law 90-
396. 90th Congress, H.R. 6279, July 11, 1968. The Mass Spectrometry Data Center (MSDC), a component of the BMD, is responsible for the development of the NIST/NIH/EPA Mass Spectral
Library, the NIST Peptide Databases, the NIST Tandem Library
(http://chemdata.nist.gov/dokuwiki/doku.php?id=chemdata:start), and the NIST
Tandem Glycan Library
(http://chemdata.nist.gov/dokuwiki/doku.php?id=chemdata:glycanlibrary). To facilitate glycan analysis, MSDC needs to acquire an instrument capable of fluorescent quantitative detection of labeled glycans.
II. SCOPE
The contractor shall provide a LC instrument with fluorescence detector meeting the specifications listed below with a one-year warranty, and four years of option year maintenance after the expiration of the warranty for the instrument.
III. SYSTEM REQUIREMENTS
The instrument must be composed of a high pressure mixing binary pump, thermostatted autosampler, thermostatted column compartment, and fluorescence detector with the features as follows:
A. GENERAL
• The LC instrument with fluorescence detector must be able to be integrated with a
Thermo Fisher Orbitrap Fusion LUMOS mass spectrometer. Integrated means the LC with detector must be completely controlled by a single mass spectrometer computer where the mass spectrometer and LC methods are created and executed in a single method file and LC method parameters are recorded in the data files.
• In addition, the LC instrument with fluorescence detector must be able to be utilized on other brands of mass spectrometers that MSDC may acquire in the future. In this case, the LC instrument with fluorescence detector does not need to be integrated and can operate on a separate computer from the mass spectrometer, but the LC must be able to signal the mass spectrometer to begin data collection.
• The instrument must include a wheeled cart to allow the instrument to be moved between mass spectrometers.
• The instrument must have backpressure capability of 1034 bar for the entire system at a flow rate of up to 5 mL/min.
http://chemdata.nist.gov/dokuwiki/doku.php?id=chemdata:start http://chemdata.nist.gov/dokuwiki/doku.php?id=chemdata:glycanlibrary
• The entire flow path must be fully biocompatible (pH range 2–12, buffer and/or chloride concentration up to 1 mol/L)
• The instrument must have a ceramic rotor seal in the injection valve that can withstand 1034 bar operational pressures.
• The instrument must have 0.1mm ID biocompatible tubing to minimize dispersion.
B. PUMP
• The pump must be able to generate flow from 0.001–8 mL/min, in 1 μL/min increments with flow accuracy of +/- 0.1% and flow precision of <0.05% RSD or
<0.01 min SD, whichever is greater.
• The pump must possess six solvent lines with built-in solvent degassing for two channels.
• The pump must be able to be operated from 20–1034 bar with flow rates up to 5 mL/min and up to 800 bar with a flow rate above 5 mL/min.
• The pump must possess proportioning precision of <0.15% SD and proportioning accuracy of +/- 0.5% (of full-scale).
C. COLUMN COMPARTMENT
• The instrument must possess an integrated temperature-controlled column compartment that can operate from 5–120 °C in 0.1 °C increments.
D. AUTOSAMPLER
• The instrument must include an autosampler with an injection volume range from
0.01–25 μL with injection precision of <0.25% area RSD for 1 μL.
• The autosampler must utilize a split loop injection.
• The autosampler must be able to accommodate a total of 54 × 12 mm OD vials autosampler vials.
• The autosampler must be able to maintain the sample compartment temperature in the range of 4–40 °C.
E. FLUORESCENCE DETECTOR
• The instrument must include a fluorescence detector that can acquire data at up to
200 Hz.
• The fluorescence detector must possess a 2 µL, biocompatible flow cell.
IV. DELIVERY
The instrument shall be delivered within 30 calendar days to the NIST Gaithersburg, MD campus.
V. INSTALLATION
The Contractor shall install the LC instrument with fluorescence detector at NIST Gaithersburg, MD. Installation shall include, at a minimum, uncrating/unpacking of all equipment, set-up and hook-up of the instrument, start-up, demonstration of specifications, and removal of all trash.
The Contractor shall demonstrate that the instrument is running in accordance with the System
Requirements. The contractor shall provide samples of standards for on-site demonstration of the performance of the LC instrument for the specifications listed. Installation shall take place between 8:30 am and 5:00 pm Eastern Time, Monday through Friday, except Federal Holidays
(hereinafter “normal duty hours”) and shall be coordinated with the NIST Technical Point of
Contact (TPOC). Installation shall be completed within 90 calendar days of award.
VI. WARRANTY
The contractor shall warrant the instrument for a period of one year, to commence after delivery, installation, training, and acceptance of the instrument by the government. The warranty shall include unlimited repair visits in the event of instrument failure during normal duty hours. The contractor shall cover expenses required for emergency repairs, including parts, labor, travel, and per diem.
VII. MAINTENANCE (OPTIONS)
The Contractor shall provide four (4) one-year options for preventative maintenance services at
NIST Gaithersburg, MD after expiration of the one-year warranty. The service shall include one preventative maintenance visit per year to ensure the instrument is performing to the manufacturer’s original specifications. Maintenance shall include all parts, labor, travel, remote support that provides troubleshooting information for identifying the source of technical problems with the instrument, and unlimited emergency repair visits within a week after the initial call. The Contractor shall provide consumables and supplies required for repair, including normal wear and tear items such as liners, seals, lamps, and tubing when needed.
1. The contractor shall respond to an unlimited number of requests for service calls in the event of instrument failure. The contractor shall respond to service calls during normal duty hours.
2. The contractor shall repair/replace all parts required for the operation of the instrument.
3. The contractor shall provide maintenance and repair by factory-trained technicians using parts, supplies, and consumables authorized for use by the manufacturer.
