STATEMENT OF NEED (SON)

1.0 TITLE

CyTOF XT/Hyperion XTi Imaging System Upgrade

2.0 BACKGROUND

The Flow Cytometry Center (FCC) is a core facility for use by employees of the National Institute of

Environmental Health Sciences (NIEHS). The FCC provides researchers with access to a variety of flow cytometric techniques to analyze cells at the single cell level and currently has over 160 investigators who use our services. The mass cytometry analyzes individual cells labeled with stable earth metal isotopes using state-of-the-art inductively coupled plasma time-of-flight technology.

With more than 120 detection channels, the mass cytometry has the capability to simultaneously resolve multiple cellular characteristics per cell at high acquisition rates, thus maximizing the information that is achieved from a single sample to generate high-resolution phenotypic and functional profiles of cells from normal and diseased states. This high parameter analysis can be accomplished both that the single cell and tissue level via mass cytometry. The CyTOF XT/Hyperion

XTi Imaging System upgrade for the FCC for NIEHS investigators to take advantage of the latest technical advances in mass cytometry.

4.0 PRODUCT FEATURES/SALIENT CHARACTERISTICS

The Flow Cytometry Center (FCC) requests the purchase of a CyTOF XT/Hyperion XTi Imaging

System Upgrade located in the Flow Cytometry Center at NIEHS. This upgrade will allow investigators to take advantage of the latest advances in mass cytometry including a larger region of interest (ROI) that can be captured in tissue sections, along with the ability to analyze the entire slide in “preview mode” to determine the best region of interest all while using less argon. This upgrade retains all the current features of our Helios mass cytometer and Hyperion imaging system including the ability to analyze over 40 individual cellular characteristics at the single cell level at high acquisition rates in both suspension and in tissue samples.

The following product features/characteristics are required for this requirement:

• Hyperion to Hyperion XTi™

• Automated temperature control

• Clog detection and auto clog removal

• Ability to run 23 hours with auto-shutdown

• 25% less argon consumption.

• New modes including Preview/Slide mode and Tissue mode

• Faster ablation: 800hr (2mm2/hr) vs 200hr (0.6mm2/hr)

• Improved MCD Viewer allowing pixel-based clustering analysis of images

• Larger region of interest size (ROI 10 mm2 vs 1.6 mm2; over 6X greater)
