About this file
This Statement of Need (SON) details a procurement for a Dymo200 C-trap instrument for the Structural Biology Core at the National Institute of Environmental Health Sciences (NIEHS). The document specifies a firm fixed-price purchase order for a specialized scientific instrument with specific technical requirements, including 2 colors confocal fluorescence, 2 dynamic optical traps, dual force detection, automated microfluidics, an optical table, a workstation rack with three screens, and associated peripherals. The C-trap will enable advanced research capabilities in studying DNA and RNA binding proteins, protein folding, and molecular interactions at a single-molecule level, with capabilities for live visualization, force measurement, and molecular tracking.
Delivery is specified for the National Institutes of Health, located at 111 TW Alexander Dr in Research Triangle Park, Durham, North Carolina, with a 10-week delivery timeline after purchase order award. The procurement includes a 36-month warranty and initial set-up training. The instrument represents a new technology not currently available at NIEHS, with broad applicability for structural biology research, including measuring protein binding events, tracking protein movement, detecting interaction partners, and observing protein condensate formation.

STATEMENT OF NEED (SON)
1.0 TITLE
Dymo200 C-trap for Structural Biology Core
2.0 BACKGROUND
The Structural Biology Core is a core facility at the NIEHS that facilitates structural and biophysical characterization of biological samples using techniques such as X-ray crystallography, chromatography, differential scanning fluorimetry, light scattering and isothermal calorimetry (ITC).
The addition of the C-trap would provide crucial information regarding real-time function of biological systems currently being studied at the NIEHS. This instrument provides for live visualization, manipulation, and force measurement at the smallest molecular scale, and single-molecule imaging and base-pair resolution measurements of the interactions between biomolecules.
This innovative out-of-the-box technology facilitates direct measurement of 1) frequency and duration of protein binding events, on- and off-rates, high precision binding location to target 2) The tracking of proteins in real-time to measure diffusion coefficients and translocation rates on DNA or
RNA, 3) Colocalization and recruitment: detection of multiple interaction partners, subunits of protein complexes, or target sequences, and their order of assembly, and 4) Condensates formation and properties: Observe proteins forming condensates with DNA or RNA and how condensates change the behavior of the DNA. This instrument has broad applicability to the study of DNA and
RNA binding proteins, protein folding, phase separation, mechanobiology as well as cytoskeletal structure and transport. This technology currently does not exist at the NIEHS.
3.0 TYPE OF ORDER
This is a:
• Firm Fixed-Price Purchase Order
• Severable
4.0 PRODUCT FEATURES/SALIENT CHARACTERISTICS
Fragmentation-based molecular annotations are the gold standard in high-confidence biomarker identification using mass spectrometry. The new imaging parallel reaction monitoring parallel accumulation serial fragmentation (iprm-PASEF) workflow integrates CCS-enabled MALDI Imaging of lipids with a targeted MS/MS imaging approach that allows spatial fragmentation and analysis of multiple targets in a single acquisition. The workflow includes a fully integrated pipeline within our software ecosystem to generate accurate identifications with spatial fidelity.
The following product features/characteristics are required for this requirement:
• C-Trap with: 2 colors confocal fluorescence (561, 638 nm)
• 2 dynamic optical traps
• Dual force detection (x,y)
• Automated microfluidics
• Optical Table
• Workstation rack with three (3) screens
• Keyboard, mouse and joystick
