About this file
This document is a Statement of Need (SON) for the acquisition of a 600 MHz Bruker Cryogenic Probe, Cryogenic Platform, and Associated Helium Compressor by the National Institutes of Health (NIH), National Library of Medicine (NLM). The purpose is to upgrade the existing NMR spectrometer capabilities to allow for the study of proteins at lower concentrations. The required equipment must be 100% compatible with the existing Bruker console and magnet system. Key features include a 1H optimized 5mm triple resonance TCI H&F cryoprobe, a Cryo Cooling Unit/5 (CU/5) cryoplatform system, and an associated helium compressor. The equipment must interface with Bruker software and be compatible with the lab's existing infrastructure.
This SON is related to a pre-solicitation (Solicitation Number: 75N97024Q00061) issued by the Department of Health and Human Services, National Institutes of Health, National Institute of Environmental Health Sciences. The applicable NAICS code is 334516 with a size standard of 1,000 employees. Interested vendors are requested to submit capability statements addressing their qualifications and ability to provide the required equipment by July 26, 2024. The government will use the information gathered to determine the acquisition strategy, which may include a small business set-aside.

Statement of Need NHLBI Office of Intramural Management (OIM) Acquisition and Logistics Branch (ALB)
Project Title 600 MHz Bruker Cryogenic Probe, Cryogenic Platform and Associated Helium Compressor
Statement of Need/Purpose and Objectives The addition of a 600 MHz Bruker 1H optimized 5 mm triple resonance TCI H&F cryoprobe with accompanying cryoplatform package will provide new capabilities, such as improved dynamic range for signal detection, allowing the laboratory to study proteins at low concentrations (20-60 uM), which are currently inaccessible to the room temperature NMR probeheads available on the instrument. At present, the laboratory’s allowed time on existing cryoprobe equipment is shared with the NHLBI Laboratory of Molecular Biophysics and instrument time shared among 6 postdoctoral fellows and 2 staff scientists is at a premium. The addition of a second cryoprobe will allow the Structural Bioinformatics Lab significantly greater access to this critical tool required to advance the lab’s research mission. Access to high-sensitivity, high field NMR spectrometer time is crucial for the lab section to fulfill its mission to investigate the biophysical properties of fold-switching proteins that are identified by our sequence-based predictions.
Product Specifications/Features/Salient Characteristics
The Structural Bioinformatics Laboratory (NLM) is requesting the purchase of an upgrade from a Bruker room temperature probe to a cryogenic probe system capable of acquiring data at least a 5-fold higher signal to noise than the existing room temperature probe, thereby allowing for the study of proteins that would not otherwise be possible due to solubility challenges. This upgrade MUST be 100% compatible with the existing, proprietary Bruker console equipment and magnet system. Without any modification, the upgraded 600 MHz cryoprobe and cryoplatform system must have these capabilities and be installed by the vendor:
1H optimized 5mm triple resonance TCI H&F CryoProbe should be:
Capable of 1H or 19F observation with 13C and/or 15N decoupling (triple or double) Capable of 13C observation with 1H and high sensitivity on 13C Probe should include helium-cooled preamplifiers for 1H & 19F, 13C, 15N and 2H and the following:
- 2H lock
- Z-gradient with 6 G/A*cm
- Automated Tuning & Matching (2G ATM)
- VT range: -40°C to +150°C
- VT gas: Nitrogen
- Automatic probe recognition by Bruker software (PICS)
CU/5 based He CryoPlatform basic system supporting HR NMR He CryoProbes should include:
- Cryo Cooling Unit/5 (CU/5)
- Utilize either a water cooled indoor He-compressor OR and indoor air-cooled He compressor
- Cooling water monitoring kit
- 6 meter indoor He-flexline set with sound insulation
- Transferline support set to absorb vibrations
- Mounting system for He CryoProbe
- He gas bottle adaptor with 10 meter flexible line
All equipment should also have the following capabilities:
-The ability to connect to existing Bruker software and console hardware in the lab.
-The new cryogenic probe must be able to interface with the Bruker Cryopanel probe monitoring software, allowing for automated cooldown and warmup.
-Seamless compatibility with a Bruker air temperature controller -Compatibility and remote control of Bruker supplied Helium compressor -The new cryogenic probe must fit the Bruker provided shimset and allow for proper sample lift operations and spinning in our superconducting magnet without modification.
-Communicate with a Bruker console, such that automated sample eject and controlled warm-up take place without damaging the probe, in the event of an emergency-triggered cryoplatform warm-up cycle.
