STATEMENT OF NEED (SON)

SS-75N91024Q00015 1

1.0 TITLE

Procurement of One (1) Brand Name or Equal Nanion Orbit Mini Complete Lipid Bilayer Recording Station

2.0 BACKGROUND

The U.S. Department of Health and Human Services (DHHS), National Institutes of Health (NIH), National Cancer Institute (NCI), Center for Cancer Research (CCR), Laboratory of Cell Biology (LCB), Crystallography Section requires the procurement of a lipid bilayer recording station. The transport of materials into and out of a cell that is surrounded by a lipid bilayer is enabled by membrane-integrated protein molecules. However, cytotoxic compounds (i.e. drugs used in chemotherapy) can, at times, pass the lipid bilayer without the need of active transport. Hydrophobic molecules can penetrate the bilayer by simply being sufficiently soluble in the very hydrophobic environment created by lipids. To protect a cell against cytotoxic compounds coming from the outside or perhaps even toxins generated on the inside in metabolic processes, membrane bound molecules (transporters) exist, which recognize those xenobiotics, bind and pump them out to the extracellular space. While the static 3D structure of many of those membrane bound efflux pumps is known, we need to understand the actual dynamic nature when the membrane efflux pumps recognize a substrate and initiate transport.

To this end, we propose the purchase of a Lipid Bilayer recording station to measure changes in electrical signals that are typical for transport events. This allows quantitative and qualitative studies of small or large, positively, or negatively charged molecules as they are moved across the membrane.

3.0 TYPE OF ORDER

This shall be a Firm Fixed-Price Purchase Order.

4.0 PURCHASE ORDER REQUIREMENTS

4.1 PRODUCT FEATURES/SALIENT CHARACTERISTICS

The following product features/characteristics are required for this requirement:

• A complete lipid bilayer recording station with four channels. A fully integrated system will save cost and valuable lab space.

• The sample must be held at a constant temperature as all reaction rates are highly sensitive to the thermal energy (temperature) of the system. The instrument needs to be equipped with a temperature control system to no only hold the temperature at the desired point but allow to set the it to higher or lower fixed temperature points than the current room temperature. This should be done with a water cooled Peltier technology which is highly efficient as it does not require large external heat baths with slow response times.

• The instrument needs to be equipped with an OPTO kit for parallel readout of electrical and optical signals. For this purpose, we require translucent recording substrates to hold the samples with a set of bores with different diameters to accommodate different sample volumes.

• The system must come with a computer (laptop) with appropriate software to run the experiment and to analyze the results.

SS-75N91024Q00015 2

• Remote technical support shall be required as well as an on-site training session that will speed up the training of the users to obtain quality data that can advance our understanding of the lipid-bound molecules and lead to publishable results.

• A service plans shall be setup to ensure the proper functioning of this recording station over the period of 1 year. This should include the free replacement of the operational amplifier if required, free spare parts and firmware upgrades for maximal performance.

4.2 WARRANTY

In addition to the Implied warranty contained in FAR Clause 52.212-4, the Contractor is encouraged to provide express warranties. At a minimum, the Contractor shall provide a 12-month warranty from date of final installation on all equipment and associated items provided under, or as a part of this purchase order.

The warranty shall include repair and or replacement of parts, including travel, labor, and materials, at no additional cost to the Government. The warranty shall not exclude or limit the Governments rights for an Implied warranty as established in the terms and conditions of clause 52.212-4.
