About this file

This is a Specifications document for the State of California related to Trimedlure Plugs (Contract P25-1024), which are essential components for Jackson Traps used in European Fruit Fly (EFF) detection and eradication programs throughout the state. The specifications detail the technical requirements and performance standards for the plugs, including their chemical composition, dimensional tolerances, and functional characteristics necessary for effective trap operation in statewide pest management initiatives.

The document establishes precise product specifications to ensure consistency and efficacy across all Jackson Traps deployed in California's EFF control program. Key specifications address the plug's lure potency, stability, shelf life, and compatibility with trap systems, ensuring that only products meeting strict quality standards are procured. Bidders must supply Trimedlure Plugs that comply with all outlined technical parameters and undergo appropriate testing and validation to confirm adherence to specifications before delivery to state agencies managing the eradication program.

Following field aging of production-lot dispensers, for 1 and 6 weeks, the number of Medflies captured shall be at least 50 percent of that caught by standard freshly baited Richmond cotton Vllick baited with

2.0 milliliter of the same bulk trimedlure as used for the dispensers .

. 1 .

Only those products, which have been tested and approved by either CDFA or USDA for use in the Mediterranean fruit fly control program, will be considered for award.

3. TESTING PROCEDURES FOR SOLID DISPENSERS

General-The Contractor is responsible for the performance of all sampling and testing as specified herein. Except as otherwise specified, the Contractor may utilize his own or any other testing facilities and services acceptable to the State. Records of the examination and tests shall be kept complete and available to the State.

3.1 Emission rate as determined by weight loss at constant temperature:

1. Release rates are determined by weight loss in an incubator oven, Thelco Model 2, Precision Scientific, Chicago. The air is admitted to the oven through six inlet ports but is not directed within or out of the oven. Six holes are drilled through the oven wall to function as exit ports to release the air. (J. Econ. Entomol. 82:860-867, 1989).

2. Two replicate plugs of each treatment are unwrapped and left to equilibrate overnight at room temperature.

3. In the morning, the plugs (no more than 8) are then placed on the shelf in the oven which has been pre-set to 28.0 plus or minus 0.5°C and into which air is flowing at the rate of 100 ml/min. through each of the six inlet ports.

4. After 1 hour of equilibration in the oven, the plugs are removed and weighed to the nearest 0.02 mg and returned to the oven.

5. The plugs are reweighed after each of three 2-hour intervals; the average weight loss/hour over the 6 hours is considered to be the emission rate.

Emission rate can also be detennined using the NMRAL Method PRO105 (See 4.0)

3.2 Trimedlure (TML) Content Measurement:

The quantity of trimedlure in each dispenser is measured by gas chromatography on solvent extracts.

The analyses for TML content are conducted on a gas chromatograph equipped with an automatic sampler and a 30m, SPB-1 (non-polar) wide-bore (0.75 mm i.d.) capillary column (Supelco, Bellefonte, PA.).

1. Single polymeric plugs are weighed, measured (length and diameter) and then dropped into a measured volume of 50/50 acetone/hexane. Two replicates of each treatment are analyzed. The volume of solvent used for extraction varies from 100 to 800 ml depending on the expected content; then a dilution is made to achie\e a concentration about 1 ug/ul.

2. The plugs are allowed to leach at room temperature for at least 48 hours with intermittent mixing prior to analysis.

3. A solution crystalline TML-C isomer of known concentration is used as the external standard; the concentration should be in the same range as the samples, i.e. about 1 ug/ul.

4. Aliquots of samples and standards are measured for analysis. Using an auto sampler,

- 2 -

1 uL injections are analyzed following these chromatographic conditions: Injector temperature - 225°C; Detector temperature - 250°C; Temperature program - initial temperature of 100 °C for 2 minutes, ramp at 10°C/minute to 250°C and hold at 250°C for 6 minutes. Total run time is 23 minutes.

5. The TML concentration is determined by comparison of the combined area of the 2 TML peaks (ca. 7 minute retention times) in the samples with the area of the single TML-C peak in the standards which bracket the samples. The total TML content of the plugs is then determined using the extraction volume in the calculation.

The GC analysis on the wide-bore column is used also to calculate the percentage of the TML which is C isomer; the first GC peak is the other trans (and many of the cis) isomers while the second peak is TML-C. The TML-C concentration is the area of the 2nd peak x 100 / total area of the 2 peaks.

NMRAL Method PRO105 is another way to measure Trimedlure content.

4. ALTERNATE TESTING PROCEDURES FOR SOLID DISPENSERS (NMRAL METHOD

PRO105.

4.4 Procedure for testing emission rate:

1. Unwrap dispenser, record the color and odor of the plugs and whether or not there are white crystals on the surface. Using double scale caliper measure the length and diameter in millimeters (mm) and record.

2. Using an. analytical balance, weigh the plug to the nearest 0.0001 grams and record the weight. Leave the plug to equilibrate overnight at room temperature.

3. Connect the vessels to the water circulator in a series. Set the water temperature to

28.0 plus or minus 0.5°C.
