About this file

This document is a Statement of Work (SOW) for a federal contract opportunity to provide sequencing services for pooled cDNA libraries using the Illumina NovaSeq X Plus instrument. The USDA Agricultural Research Service (ARS) requires these sequencing services to advance research under an in-house project.

The key details are: The contractor will conduct NovaSeq X Plus 10B (2x150bp) sequencing of pooled cDNA libraries provided by the government. The contractor will perform library quality control analysis and sequence libraries that pass QC. Demultiplexed raw sequencing data in FASTQ format will be delivered within 30 days of QC approval. The sequencing services will be conducted for a minimum of 1 lane and a maximum of 15 lanes. The place of service and acceptance is Ithaca, NY. This is a total small business set-aside with NAICS code 541714 and 1,000-employee size standard. The evaluation will be based on the Lowest Priced Technically Acceptable (LPTA) approach.

Statement of Work (SOW) Project: Sequencing of DNA libraries using NovaSeq X Plus instrument.

GENERAL INFORMATION

1.0 Scope of Work:

The USDA-ARS Plant, Soil, and Nutrition Research Unit in Ithaca, New York requires NovaSeq X Plus 10B (2x150bp) sequencing services on pooled cDNA libraries.

2.0 Background:

The requirement is the completion of nucleic acid sequencing of pooled cDNA libraries to advance research efforts on Objective 1 of the in-house project 8062-21000-050-000-D.

CONTRACTOR REQUIREMENTS

3.0 Technical Requirements/Tasks:

Transcriptome Sequencing:

Conduct NovaSeq X Plus 10B (2x150bp) sequencing of pooled cDNA libraries. Illumina compatible libraries will be sent to the service provider along with index information for each sample in the library. Service provider will conduct Library QC analysis including library template size distribution using a TapeStation instrument and library template quantification using qPCR. Library quality control results will be delivered by the service provider. Libraries that pass quality control will be sequenced. Each library will be sequenced on its own lane of NovaSeq X Plus instrument using the 300 cycle reagents on the 10B flow cell. Demultiplexed raw data in fastq format will be delivered by the service provider. Sequencing services will be conducted for a minimum of 1 lane and a maximum of 15 lanes.

4.0 Government Furnished:

Nucleic acid samples consisting of pooled cDNA libraries will be provided. Library index information will be provided.

5.0 Deliverables / Schedule:

Service provider is expected to provide QC results and analysis for each library within 30 days of sample receipt. Libraries that pass QC will automatically proceed to library sequencing and libraries that fail QC will await client approval prior to sequencing. All raw sequencing data will be demultiplexed and delivered in the fastq format. Delivery of raw sequencing data will be made through a secure file transfer protocol and is expected within 30 days of QC approval. Data will be stored on the service provider’s servers for 3 months from the date of completion.
